The major Fc receptor in blood has a phosphatidylinositol anchor and is deficient in paroxysmal nocturnal haemoglobinuria

Abstract: Fc receptors on phagocytic cells in the blood mediate binding and clearance of immune complexes, phagocytosis of antibody-opsonized microorganisms, and potently trigger effector functions, including superoxide anion production and antibody-dependent cellular cytotoxicity. The Fc receptor type III (Fc gamma R III, CD 16), present in 135,000 sites per cell 1 on neutrophils and accounting for most of FcR in blood, unexpectedly has a phosphatidylinositol glycan (PIG) membrane anchor. Deficiency of Fc gamma R III i… Show more

“…This protein is also a Pltailed protein like the Fc?'RIII. The lowered expression of these two Pldinked proteins in CML seems to follow the trend that is observed with respect to the decreased expression of Pl-linked proteins, leukocyte alkaline phosphatase, acetylcholine esterase, FcyRIII and dccay acceleration factor in PNH patients [13][14][15][16]. In the latter disorder however, the lowered expression of these proteins is believed to be associ~te£ -,,.…”

“…The anti-CD16 nonblocking monoclonal antibody (mAb) 214.1 (murine IgG1) was a generous gift from Dr. Howard Fleit (State University of New York, Long Island). The anti-CD16 adhesion blockade mAb CLBFcgran-1 (murine IgG2a) was purified in house from hybridomas as previously described (20). Cleavage of CLBFcgran-1 into Fab fragments was done by Lampire (Pipersville, PA).…”

“…However, relatively little is known about the induction and the mechanism(s) of such processes. Although Fc␥RIIIb is GPI anchored to the cell membrane (18), several groups have established surprisingly that it can transduce signals by itself (19 -21). In contrast, others have stated that Fc␥RIIIb must cooperate with one or several neighboring transmembrane partners.…”
