The m6A(m)-independent role of FTO in regulating WNT signaling pathways

Abstract: FTO and ALKBH5 are the two enzymes responsible for mRNA demethylation. Hence, the functional study of FTO has been focused on its mechanistic role in dynamic mRNA modification, and how this post-transcriptional regulation modulates signaling pathways. Here, we report that the functional landscape of FTO is largely associated with WNT signaling pathways but in a manner that is independent of its enzymatic activity. Re-analyses of public datasets identified the bifurcation of canonical and noncanonical WNT pathw… Show more

“…Given the growing evidences that m6A is associated with Wnt signaling 5 , 6 , while recognizing that Wnt/β-catenin plays an inevitable role in GBM 7 9 , there may be possible mechanisms for their yet to be elucidated interplay in the pathogenesis of GBM. Concerning the functional regulation of m6A, the fine-tuning and interactions between the m6A and long non-coding RNAs (lncRNA) axis have been considered.…”

“…WTAP has been reported to be associated with a number of signaling pathways, such as TGFβ (Li et al, 2020b), hippo (Hu et al, 2020), NF-κB , and Hedgehog pathways (Wei et al, 2022). FTO is associated with various signaling pathways, for example, PKA/CREB (Hu et al, 2022), TNF-α (Li et al, 2022b), ERK (Xiao et al, 2022b), WNT (Kim et al, 2022) and JAK2/ STAT3 pathways (Shen et al, 2021). ALKBH5 is involved in many signaling pathways, including WNT (Lin et al, 2022), PTEN/AKT (He et al, 2021), NF-κB (Qu et al, 2022), AKT (Wang et al, 2020).…”

“…m6A methylation modification plays an important role in maintaining stem cell metabolism [ 29 ], immune homeostasis [ 30 ], tumor immune microenvironment [ 31 ], immune suppression and escape [ 32 ], and other biological regulations. m6A methylation modification can also affect cell differentiation and proliferation by affecting the expression of transcription factors [ 33 , 34 ]. m6A methylation is modified by altering RNA folding and structure, which enhances mRNA export and translation [ 10 , 35 , 36 ].…”
