BackgroundThe Ly‐6 family has many members, including Ly‐6C and Ly‐6G. A previous study suggested that the anti‐Ly‐6G antibody, RB6‐8C5, may react with Ly‐6Chi murine bone marrow (BM) cells. This finding has been interpreted as cross‐reactivity of RB6‐8C5 with the Ly‐6C antigen, and has been generalized to many hematopoietic cell types, using the terminology Ly‐6G/C. The present study was undertaken to determine whether anti‐Ly‐6G antibodies truly cross‐react with the Ly‐6C antigen on multiple hematopoietic cell types.MethodsSplenocytes, thymocytes, and BM cells obtained from Ly‐6.1 and Ly‐6.2 strains of mice were stained with a variety of antibodies to Ly‐6C and Ly‐6G. Flow cytometric analysis was performed on these populations.ResultsEvaluation of anti‐Ly‐6C and anti‐Ly‐6G staining showed only Ly‐6C expression and no Ly‐6G expression on subsets of splenic T and B cells and thymocytes from Ly‐6.1 and Ly‐6.2 mice. Bone marrow cells were identified that express both Ly‐6G and Ly‐6C; no Ly‐6G+Ly‐6C− populations were seen.ConclusionsMultiple Ly‐6C+ hematopoietic cell populations were identified that do not stain with anti‐Ly‐6G antibodies. This calls into question the use of the Ly‐6G/C nomenclature and suggests that epitopes recognized by anti‐Ly‐6G antibodies should simply be designated Ly‐6G. © 2004 Wiley‐Liss, Inc.

SummaryEarly in ontogeny thymocytes have a surface marker phenotype that resembles activated mature T cells but they lack expression of the T cell receptor (TCR) complex. We have made preparations of day 14/15 triple negative fetal thymocytes that exhibit the activated T lymphocyte markers CD25, intercdlular adhesion molecule 1, Ly-6A/E, CD44, and heat stable antigen and are rapidly proliferating as evidenced by flow cytometric examination of BrdU incorporation. We found that binding activities of the gene regulators nuclear factor (NF)-gB, the NF-gB p50 homodimer complex, nuclear factor of activated T cells (NF-AT), oct-l, oct-2, activator protein 1 (AP-1), and serum response factor (SRF), are all present in these early thymocytes. Whereas the octamer factors and SRF persist during ontogeny, NF-gB, NF-AT, and AP-1 decrease and are undetectable in the adult thymus. Transfection of disaggregated thymocytes by dectroporation or intact thymic lobes by gold-particle bombardment revealed that reporter constructs for NF-gB, NF-AT, AP-1, octamer factors and, to a small extent, the TCR-ot enhancer were active in early thymocyte development. We rigorously diminated the possibility that these transcriptional events were due to minor populations of TCR + cells by showing that these reporter constructs were also active in recombinase activating gene (RAG) -/. thymocytes that are incapable of completing TCR gene rearrangement, and predominantly contain cells that have an activated phenotype. Thus, transcriptional events that are usually triggered by antigen stimulation in mature T cells take place early in thymic ontogeny in the absence of the TCR. Our analysis suggests that there are striking regulatory similarities but also important differences between the activation processes that take place in antigen-stimulated mature T cells and thymic progenitor cells.

SUMMARYWe recently described the identity of Ihe small cell lung cancer (SCI.C) cluster-w4 antigen and the human B cell ditlcrentiation marker CD24, a glycosylphosphatidylinosilol (GPI)-anchored, highly glycosylalcd surface molecule ofonly 31 -35 amino acids [15]. The specificities of three anti-cluster-w4 and of eleven anti-CD24 MoAbs have been investigated with respect to their binding capacity lo the protein core of cluster-w4/CD24 antigen. Four overlapping peptides spanning ihis protein core were synthesized. MoAbs shown to bind to two overlapping peptides by antibody binding inhibition using the cluster-w4/CD24-positive SCLC cell line SW2 and by direct peptide binding detecied in an ELISA were investigated in more detail. To determine the exact epitopes recognized by these MoAbs. an epitope mapping assay using peptides synthesized onto polyethylene pins was established. The three anti-cluster-w4 MoAbs SWA 11, SWA2I and SWA22 and the anti-CD24 MoAbs OKB2 and ALB9 recognized the same short leucine-alanine-proline (LAP) sequence in an area without potential glycosytation sites close lo the GPI anchor oi the protein core of the cluster-w4/CD24 antigen.Keywords epitope mapping monoclonal aniibody SWA 11 clusler-w4 antigen CD24 antigen small cell lung cancer
