Through a mutualistic relationship with woody plant roots, ectomycorrhizal fungi provide growth-limiting nutrients, including inorganic phosphate (Pi), to their host. Reciprocal trades occur at the Hartig net, which is the symbiotic interface of ectomycorrhizas where the two partners are symplasmically isolated. Fungal Pi must be exported to the symbiotic interface, but the proteins facilitating this transfer are unknown. In the present study, we combined transcriptomic, microscopy, whole plant physiology, X-ray fluorescence mapping, P labeling and fungal genetic approaches to unravel the role of HcPT2, a fungal Pi transporter, during the Hebeloma cylindrosporum-Pinus pinaster ectomycorrhizal association. We localized HcPT2 in the extra-radical hyphae and the Hartig net and demonstrated its determinant role for both the establishment of ectomycorrhizas and Pi allocation towards P. pinaster. We showed that the host plant induces HcPT2 expression and that the artificial overexpression of HcPT2 is sufficient to significantly enhance Pi export towards the central cylinder. Together, our results reveal that HcPT2 plays an important role in ectomycorrhizal symbiosis, affecting both Pi influx in the mycelium and efflux towards roots under the control of P. pinaster.

Interplay of complex decay processes after argon 1s ionization

Complex decay pathways involving radiative and nonradiative relaxation after deep core-level ionization in argon are disentangled by a unique combination of several synchrotron radiation-based spectroscopic techniques. In particular, by comparing the results obtained from electron-ion coincidence, photon-ion coincidence, and x-ray emission measurements, we are able to distinguish the final ionic states produced in the cascade decay involving Kα and Kβ radiative decay and final ionic states produced by nonradiative cascade decay. High-resolution Auger electron spectroscopy is then used as a complementary tool to identify the LMM transitions contributing to the cascade decay. Ab initio calculations are performed to identify the electronic states involved in the LMM decay.

The absence of an extensive series of Sc K-edge X-ray absorption near-edge structure spectroscopy (XANES) reference spectra and the scarcity of direct structural data on Sc are major hurdles to develop our understanding of Sc chemistry. However, this first step is essential to develop new Sc-based applications and to better understand the formation of Sc deposits. Here, we present a detailed comparative study of Sc K-edge XANES spectra of three Sc-bearing compounds: a garnet (CaScSiO), an oxide (ScO) and a phosphate (ScPO·2HO). First-principles calculations have been performed to interpret the origin of the K pre-edge spectral features. We demonstrate the validity of our approach by reproducing satisfyingly the experimental spectra. The densities of states projected on the absorbing Sc atom and its first neighbours give the possibility to interpret the position and intensity of the pre-edge XANES features in terms of Sc local environment. The pre-edge features provide information on p-d mixing of the absorber orbitals giving clues on the centrosymmetry of the site and on the mixing of the empty 4p orbitals of the absorber with empty 3d orbitals of the neighbours via the empty p orbitals of the ligands. We also show that these features give a first estimate of the crystal-field splitting energy (ca. 1.5 eV), inaccessible using other spectroscopic methods. Comparisons with K-edge spectra of other 3d ions from the literature reveal the specificities of the Sc pre-edge, indicating that core-hole screening is weaker than for Ti-bearing compounds. This study provides a dataset of spectral signatures and a theoretical basis for their interpretation, a requirement for future studies on Sc chemical form in synthetic and natural systems.
