The loudness dependency of the auditory evoked N1/P2-component as a predictor of the acute SSRI response in depression

Abstract: Our finding confirms the hypothesis that a strong LDAEP, indicating a low serotonergic activity, is related to a favorable response to acute SSRI treatment in depression. The LDAEP is a promising tool for the prediction of response to serotonin agonists in depression and it seems to be of clinical importance.

“…N1-peaks (50-150 ms) and P2-peaks (100-250 ms) were determined semiautomatically at the Fz-and Cz electrode (referred to linkedmastoids). The LD was calculated as linear regression slope with stimulus intensity as independent and N1/P2-amplitude as dependent variable (Gallinat et al, 2000).…”

“…For example, the LD in behaving cats was found being decreased by application of the 5-HT1a-receptor agonist 8-OH-DPAT and increased by the 5-HT2-receptor antagonist ketanserin (Juckel et al, 1997). Empirical clinical support for this hypothesis comes from studies showing that a strong LD in depressed patients is related to a favorable therapeutical outcome to serotonin agonistic agents (Gallinat et al, 2000;Hegerl and Juckel, 1993). Moreover, intraindividual changes of blood serotonin concentrations were negatively correlated with corresponding changes of the LD in patients with major depression (Hegerl et al, 1991).…”

“…In clinical studies, an increased LD has been shown in MDMA users (Tuchtenhagen et al, 2000;Croft et al, 2001) and patients with borderline personality disorder (Norra et al, 2003), indicative of a serotonergic dysfunction in these subjects. In depressed patients, a high LD before drug treatment has been associated with a favorable response to serotonergic medication (Hegerl and Juckel, 1993;Gallinat et al, 2000;Mulert et al, 2002). The observation that allelic variants of the serotonin transporter gene differ with respect to the LD further supports the hypothesis of an association between the serotonergic system and neurophysiological measures (Gallinat et al, 2003).…”

“…Depressed patients with low serotonergic activity, as evidenced by pronounced LDAEP prior to treatment, respond better to SSRI antidepressants compared to patients with high serotonergic activity, as evidenced by weak LDAEP (Gallinat et al, 2000;Hegerl et al, 2001;Lee et al, 2005;Paige et al, 1994). Although Paige et al (1995) found a similar difference in LDAEP between a small sample of responders (n=4) versus nonresponders (n=4) to an NDRI antidepressant (bupropion), more recent studies suggest that LDAEP may differentially predict clinical response to serotonergic and noradrenergic antidepressants.…”

“…Thus, studies have not found a change in LDAEP following treatment with SSRI or other antidepressants (Gallinat et al, 2000;Linka et al, 2009;Paige et al, 1994Paige et al, , 1995. Also, acute enhancement of serotonin levels by administrating an SSRI to healthy adults has not yielded consistent findings.…”
