The long non-coding RNA H19: an active player with multiple facets to sustain the hallmarks of cancer

Abstract: Cancer cells exhibit hallmarks in terms of proliferation, resistance to cell death, angiogenesis, invasion, metastasis, and genomic instability. Despite the progress in cancer research and the comprehension of tumorigenesis mechanisms, cancer remains a major issue in public health. A better understanding of the molecular factors associated with the appearance or progression of cancer may allow the development of therapeutic alternatives. Increasing data highlight the role of long noncoding RNAs in many disease… Show more

“…9 Recently, lncRNAs received much attention because of their crucial regulatory functions in various biological processes. 10,11 Moreover, a growing body of evidence suggests that lncRNAs may play complicated and crucial roles during tumorigenesis and tumor progression. [12][13][14] Many lncRNAs have turned out to be abnormally expressed in cervical cancer.…”

“…Following the same mechanism as observed for the beta-actin mRNA, IGF2BP1 also regulates the subcytoplasmic distribution of the lncRNA H19 [65]. H19 is a 2.3-kb-long, spliced and polyadenylated lncRNA that is, similarly to IGF2BP1, expressed during embryonic development and reactivated in several types of cancer [65,103,104,138]. H19 shows a diverse range of actions: it functions as a ceRNA, induces or represses the transcription of various genes, and interacts with and thus modulates the activity of different proteins like p53, thereby contributing to all hallmarks of cancer [138,139].…”

H19 shows a diverse range of actions: it functions as a ceRNA, induces or represses the transcription of various genes, and interacts with and thus modulates the activity of different proteins like p53, thereby contributing to all hallmarks of cancer [138,139]. Runge et al discovered that IGF2BP1 binds to the 3 end of H19 with high affinity and thereby targets the lncRNA to lamellipodia and perinuclear regions of proliferating mouse embryonic fibroblasts [65].…”

“…The production and signal transmission of growth-promoting factors must be fully controlled, and checkpoints must be set up at each node of cell division to ensure tissue homeostasis (cell number, tissue structure, and function). 46 In cancer cells, this balance is disrupted. SNHG16 has been shown to sustain proliferation of cancer cells in different types of cancers.…”
