The long non-coding RNA ET-20 mediates EMT by impairing desmosomes in breast cancer cells

Abstract: The vast majority of breast cancer-associated deaths are due to metastatic spread of cancer cells, a process aided by epithelial-mesenchymal transition (EMT). Mounting evidence has indicated that long non-coding RNAs (lncRNAs) also contribute to tumor progression. We report the identification of 114 novel lncRNAs that change their expression during TGFβ-induced EMT in murine breast cancer cells (referred to as EMT-associated transcripts; ETs). Of these, the ET-20 gene localizes in antisense orientation within … Show more

“…This process thus results in the impairment of intercellular junctions and enhancement of EMT. [158]. In conclusion, the regulatory activities of miRNAs and lncRNAs as post-transcriptional regulators form a complex regulatory network that controls EMT.…”

“…154 For example, impaired desmosomes were shown to promote EMT and consequent tumor progression in invasive breast cancer. 155 Besides, loss of desmosomes was observed in invasive pancreatic neuroendocrine tumors (PNET), and genetic deletion of desmoplakin enhanced tumor metastasis in the PNET mouse model. 156 Interestingly, the proper functions of desmosomes require particular posttranslational modifications on several components.…”

“…The vast majority of cancer-related deaths, such as hepatocellular carcinoma and breast cancer (Saxena et al, 2021;Gao et al, 2022), are due to metastatic spread of cancer cells, a process aided by epithelial-mesenchymal transition (EMT) (Atay, 2020;Kitz et al, 2021). EMT refers to the biological process in which epithelial cells are transformed into cells with a mesenchymal phenotype through specific programs (Zhang et al, 2022).…”
