The Local Anesthetic Bupivacaine Inhibits the Progression of Non-Small Cell Lung Cancer by Inducing Autophagy Through Akt/mTOR Signaling

Abstract: Non-small cell lung cancer (NSCLC) is a prevalent malignancy with high mortality and poor prognosis. Bupivacaine serves as a widely used local anesthetic and presents potential anti-tumor activity. Nevertheless, the function of bupivacaine in the NSCLC development remains elusive. Here, we tried to investigate the impact of bupivacaine on the NSCLC progression. Significantly, we revealed that bupivacaine was able to reduce the proliferation and induce the apoptosis of NSCLC cells. Bupivacaine could attenuate t… Show more

“…Recently, several works have described the adjuvant anticancer action of local anesthetics such as LDC [19,20], bupivacaine [21][22][23][24] and ropivacaine [25]. LDC can suppress cancer cell growth (in vitro and in vivo) through several mechanisms: regulation of epigenetic changes, promotion of pro-apoptotic pathways and regulation of ABC transporters, also preventing metastasis and angiogenesis.…”

“…Researchers found that autophagy played a vital role in tumorigenesis and development. Gu et al confirmed that bupivacaine-induced autophagy through Akt/mTOR signaling, inhibiting the progression of NSCLC (Gu et al, 2021). Lin et al proposed that high expression of miR-30a improved the prognosis of NSCLC after neoadjuvant chemotherapy by reducing autophagy caused by chemotherapy drugs (Lin et al, 2021).…”

“…Interestingly, all these screen hits are linked to different processes of autophagy. Rapamycin is a well-studied mTOR inhibitor that triggers autophagy and ubiquitin-proteasome system [29]; Bupivacaine has been reported to induce protein degradation in muscle cells [30,31] and also induce autophagy in lung cancer cells [32]; ABT199 is a BCL2 inhibitor that has the potency to regulate apoptosis and autophagy pathway [33]. These data imply that manipulating protein-specific degradation is a plausible approach to inhibit PreS2-LHBS.…”
