The Light-Harvesting Complex of Photosystem I in Chlamydomonas reinhardtii: Protein Composition, Gene Structures and Phylogenic Implications

Abstract: . In order to assign LHCI proteins in the thylakoid membranes, the PSI-LHCI supercomplex that retains all of the major LHCI proteins was purified. Seven distinct LHCI proteins were resolved from the purified supercomplex by a high-resolution SDS polyacrylamide gel electrophoresis, and their N-terminal amino acid sequences were determined. One LHCI protein (band e) was newly found, although the other six LHCI proteins corresponded to those previously reported. Genomic clones encoding these seven LHCI proteins w… Show more

“…We performed sequence alignment and phylogenetic analysis as previously described in ref. 36, displaying bootstrap values (% of 1,000 replicates) at the appropriate nodes. LHCI and LHCII protein names are after Stauber et al (17) and Minagawa and Takahashi (19), respectively.…”

“…Mass spectrometric analysis of the 2-DGEseparated Lhca protein spots confirmed this large variability and identified nine different Lhca proteins from C. reinhardtii (113). Of these nine, Lhca2 and Lhca9 were not found by N-terminal Edman amino acid sequencing (122). For the naming of the Lhca proteins we follow the nomenclature of Stauber et al (113) (Table 1).…”

“…In C. reinhardtii at least seven distinct LHCI subunits have been identified (11,120). From seven Lhca proteins, N-terminal amino acid sequences have been obtained by Edman amino acid sequencing (120,122). Separation of isolated PSI complexes from C. reinhardtii by 2-DGE revealed the presence of about 18 LHCI protein spots, suggesting an even higher variability of Lhca proteins (56).…”

“…In C. reinhardtii thylakoids, Stt7 has been found associated with Cytb 6 f, LHCII and PSI, but not with PSII (Lemeille et al 2009). Phosphorylation and migration of the PSII linker protein CP29 have also been reported to be vital for docking of P*LHCII proteins to PSI in C. reinhardtii, but the extent to which CP29 moves between the photosystems seems to depend on the experimental conditions used (Tokutsu et al 2004;Lemeille et al 2010). In higher plants, CP29 phosphorylation regulates disassembly of PSII supercomplexes under high light (Fristedt and Vener 2011).…”
