The level of DLDB/CHIP controls the activity of the LIM homeodomain protein Apterous: evidence for a functional tetramer complex in vivo

Abstract: The LIM homeodomain (LIM-HD) protein Apterous (Ap) and its cofactor DLDB/CHIP control dorso- ventral (D/V) patterning and growth of Drosophila wing. To investigate the molecular mechanisms of Ap/CHIP function we altered their relative levels of expression and generated mutants in the LIM1, LIM2 and HD domains of Ap, as well as in the LIM-interacting and self-association domains of CHIP. Using in vitro and in vivo assays we found that: (i) the levels of CHIP relative to Ap control D/V patterning; (ii) the LIM1 … Show more

“…4F). As ap is expressed in all notal cells after the second larval period (DiazBenjumea and Cohen, 1993;Rincon-Limas et al, 2000) and precedes eyg expression, we believe that inducing the activity of eyg early in notum development programmes the whole notum to develop as the eyg/toe domain, thus preventing the formation of scutellum (see Discussion). The transformations observed in clones of eyg-expressing cells (data not shown) fit well with those described with the Gal4 lines.…”

“…The tetrameric model for LIM-HD function based on studies of Ap, dLMO, and CHIP, requires two Ap molecules binding a dimer of CHIP, a process that dLmo proteins can interfere with by competing for binding to CHIP (Fernandez-Funez et al, 1998;Milan and Cohen, 1999;Rincon-Limas et al, 2000). To determine the combinations in which members of the mouse LIM-HD, Lmo, and Clim families may function in the cortex, a comparison of the spatial overlap in expression of these genes is essential.…”

“…The developing wing of Drosophila has proven a tractable system in which to study the function of complexes formed by LIM-HD proteins and their co-factors (Fernandez-Funez et al, 1998;Milan et al, 1998;Shoresh et al, 1998;Zeng et al, 1998;Milan and Cohen, 1999;Rincon-Limas et al, 2000;. The wing imaginal disc is divided into distinct lineage-restricted compartments along both the anteroposterior and dorsoventral (DV) axes.…”
