The Kinetics of Synthesis of Early Viral Proteins in KB Cells Infected with Wild-type and Transformation-defective Host-range Mutants of Human Adenovirus Type 5

Abstract: SUMMARYWe have studied the kinetics of early adenovirus type 5 (Ad5) protein synthesis during lytic infection of KB cells by wild-type (wt) and transformation-defective hostrange (hr) mutants. Proteins encoded within four early regions were studied: early region 1A (E1A: 1.5 to 4.5 map units, mu), E1B (4.5 to 11.2 mu), E2A (61.6 to 74-9 mu), and E4 (91.4 to 99.1 mu). Synthesis of E 1A products, the first to appear during wt lytic infection, was detectable within 2 h after injection, reached a peak within the n… Show more

“…The wild-type E1B protein was first clearly detected 12 h after infection and increased substantially in steady-state concentration by 18 h (Fig. 1A), in agreement with previous observations (9,58,59,65). All E1B 55-kDa proteins encoded by the mutant viruses exhibited a similar temporal pattern of accumulation, increasing in the same apparent proportion as the wild type as the late phase progressed.…”

“…In summary, our data demonstrate that wild-type-Ad5-in-fected A549 cells display almost the same pattern of expression of E1B-55kDa, E4orf6, and E4orf6/7 proteins as that previously described for infected HeLa cells (9,39,44,52). Having established the experimental conditions needed to analyze the E1B-E4 protein interaction in infected A549 cells, we next mapped the domains in E1B-55kDa required for binding to the E4orf6 protein in vivo.…”

“…Human HeLa and KB cells, as well as mouse 10T1/2 cells (47), were also cultured as described above. Cells were infected with wild-type (wt) or mutant virus at a multiplicity of 35 PFU per cell, as was described previously (48). For double infections, the viruses were premixed before infection at a multiplicity of 35 PFU per cell for each virus.…”
