For centuries, mushrooms have been appreciated as sources of food nutrients and pharmacologically important compounds useful in medicine. Yet not all the medicinal properties of mushrooms have been exploited. The above statement is more pertinent to mushrooms that are indigenous to Nigeria. There are inadequate data on the identity and medicinal properties of these wild mushrooms. Information on the ethnomedicinal uses of some mushrooms such as Pleurotus tuber-regium used for headache, stomach pain fever, cold, constipation; Lentinus squarullosus for mumps, heart diseases; Termitomyces microcarpus for gonorrhea; Calvatia cyathiformis for leucorrhea, barreness; Ganoderma lucidum for treating arthritis, neoplasia; G. resinaceum used for hyperglycemia, liver diseases (hepatoprotector); G. applanatum used as antioxidant and for diabetes had been gathered through survey. The above information is mostly obtained from traditional herbalists who in most cases will not disclose their preparation compositions. A lot of these mushrooms are obtained only in the wild. Scientific documents of the identities and medicinal properties are still scanty. Preliminary studies on some species of Temitomyces, Lenzites and Lentinus species showed that they possess appreciable antimicrobial and antioxidant properties. Moreover, molecular characterization also reveals that they are not 100% homologous with existing sequences under the same name in GenBank. It is therefore pertinent that well structured studies on their ecology, identification and medicinal uses be carried out. This will make the full exploitation of the medicinal potentials of mushrooms indigenous to Nigeria realizable.

Flammulina velutipes is known as edible and medicinal mushroom. The fungus grow on living trees of Zelkova carpinifolia and dead trees of Diospyros lotus (Stand and fallen) in Behshar forest area. These plants were found as new hosts for F. velutipes. In vitro antagonistic activity of this mushroom against Bipolaris sorokiniana, Ophiostoma ulmi, Pestalotiopsis funerea, Fusarium culmurum, and Fusarium oxysporum was evaluated on MEA as dual culture. Colonies of the fungus were white farinaceous and powdery on MEA, later turns yellow. Daily growth of colony was measured 5.36mm on MEA at 25 o C. Growth coefficient was indicated 36.64 and 90 mm Petriplate was covered up to 15 days after plating. The strongest and poorest antagonistic index have been seen against Ophiostoma ulmi and Bipolaris sorokiniana, respectively.

Orthogonal experiment was used to optimize the extraction conditions of Flammulina velutipes mycelia polysaccharides (FvP). Four independent variables (ratio of water to raw material, initial pH value, extracting temperature, and extracting time) were taken into consideration. A yield of FvP of 2.19% was obtained under an optimized condition (ratio of water to material of 50:1, initial pH value of 6.0, extracting temperature of 85℃, and extracting time of 6 h). Subsequently, antioxidative properties of FvP-2 (crude polysaccharides) and FvP-3 (deproteinized polysaccharides) prepared from F. velutipes mycelia were evaluated by monitoring the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical and hydroxyl radical scavenging activity, together with antitumor activity against the human hepatocellular carcinoma BEL-7402 cells. DPPH scavenging rate of 65.85% and hydroxyl radical scavenging rate of 71.24% were obtained at 200 μg/mL of FvP-3. The inhibition rate of BEL-7402 cells was up to 45% at 640 μg/mL of FvP-2. These results suggested that FvP possesses potent antioxidant and antitumor properties. The polysaccharide may be useful as a functional food additive and an antioxidant and antitumor agent. IntroductionMushrooms have been valued as edible and medicinal resources, and antitumor substances have been identified in many mushroom species. Polysaccharides, as one of the most important components isolated from mushrooms, have been correlated with multiple pharmacological activities, such as antioxidant, immunomodulatory, reducing blood lipid and antitumor activities. Flammulina velutipes, one of the most popular edible mushrooms, has being under a large-scale artificial cultivation and increasingly consumed in Japan and China owing to its high nutritional values and attractive taste (Smiderle et al., 2008).
