The Intrinsically Disordered Region of Coronins Fine-tunes Oligomerization and Actin Polymerization

Abstract: Coronins are highly conserved actin-binding proteins (ABPs) in the eukaryotic kingdom for polymerizing actin cytoskeleton. The biochemical activity of coronins is primarily mediated by the structural N-terminal β-propeller and the C-terminal helical coiled-coil (CC) domains, but less is known about the function of a middle nonconserved region, the “unique region (UR)”. The coronin UR is an intrinsically disordered region (IDR). Herein, we demonstrate that the low complexity of the UR is a conserved signature o… Show more

“…Notably, MoSpa2(1-338) exhibits a high degree of intrinsic disorder and is rich in charge blocks and Pi-Pi interactions, as revealed by NCPR, FCR, and Pi-Pi analysis (Appendix Fig S4B). These characteristics often play a role in facilitating multivalent interactions that contribute to molecular condensation (Han et al , 2023; Miao et al ., 2018; van der Lee et al , 2014; Vernon et al , 2018).…”

These characteristics often play a role in facilitating multivalent interactions that contribute to molecular condensation (Han et al, 2023;van der Lee et al, 2014;Vernon et al, 2018). Given MoSpa2's crucial role in structuring a nucleation centre at the growing tip in vivo, and its sequence signatures indicating surface charge block-and coiled-coil (CC) oligomerization-mediated phase separation through Pi-Pi interactions, we explored their potential for homotypic demixing across varying protein concentrations and ionic strengths.…”
