The Interrelationship of Sputum Inflammatory Markers in Patients with Chronic Bronchitis

Abstract: Many of the features of bronchial disease are believed to be caused by damage to the airways by elastase released by recruited neutrophils. There have been few studies of the mechanisms involved and the interrelationships between components of the inflammatory process. We studied secretions from patients with chronic bronchitis in the stable state. We assessed the presence of neutrophils by measuring myeloperoxidase (MPO) activity and active neutrophil elastase (NE). These results were compared with the chemoa… Show more

“…Active neutrophil elastase was measured using a chromogenic substrate specific for human neutrophil elastase, n-methoxysuccinyl-L-alanyl-L-alanyl-prolyl-L-valyl-p-nitroanilide (Sigma), as described previously. 26 Both IL8 and neutrophil elastase have been previously validated for assessment in induced sputum. 27 SP-A concentration was determined by adapting a previously described method.…”

“…These assays and their characteristics have been described in detail elsewhere [24]. a 1 AT concentration in sputum sol phase was measured by ELISA relative to a commercially available serum standard (The Binding Site Ltd., Birmingham, UK) as described previously [16] and a 1 AT in serum was measured by radial immunodiffusion (The Binding Site Ltd.).…”

“…Moreover, sputum fluid-phase in COPD shows an increase in neutrophilic and eosinophilic granulocyte proteins such as Myeloperoxidase (MPO), Human Neutrophil Lipocalin (HNL), Neutrophil Elastase (NE) and Eosinophilic Cationic Protein (ECP), Eosinophil Peoxidase (EPO) and chemoattractants like Leukotriene B-4 (LTB-4), Growth Related Oncogen-a (GRO-a), Monocyte Chemotactic Protein-1 (MCP-1) and Granulocyte Macrophages-Colony Stimulating Factor (GM-CSF) [58][59][60].…”
