The interplay of histone modifications – writers that read

Abstract: Histones are subject to a vast array of posttranslational modifications including acetylation, methylation, phosphorylation, and ubiquitylation. The writers of these modifications play important roles in normal development and their mutation or misregulation is linked with both genetic disorders and various cancers. Readers of these marks contain protein domains that allow their recruitment to chromatin. Interestingly, writers often contain domains which can read chromatin marks, allowing the reinforcement of … Show more

“…We are beginning to understand how the much greater diversity of protein methylation reactions can give rise to a greater diversity of function (1,(4)(5)(6)(7)(8). We are also learning the importance of crosstalk between protein and DNA methylation reactions (9) and among protein methylation, phosphorylation, acetylation, and ubiquitination reactions (10)(11)(12). Finally, we are discovering how alterations in protein methylation pathways can lead to human pathology, particularly in cancer (13)(14)(15).…”

“…The histone 3 (H3) tail serves a central role in epigenetic regulation primarily through alterations of lysine methylation (Zhang et al, 2015). Trimethylation of lysine 4 (H3K4me3) relates to trithorax-Group (trx-G) action (Roguev et al, 2001) and is a hallmark of active promoters (Santos-Rosa et al, 2002;Bannister and Kouzarides, 2011).…”
