The Interplay between Oncogenic Signaling Networks and Mitochondrial Dynamics

Abstract: Mitochondria are dynamic organelles that alter their organization in response to a variety of cellular cues. Mitochondria are central in many biologic processes, such as cellular bioenergetics and apoptosis, and mitochondrial network morphology can contribute to those physiologic processes. Some of the biologic processes that are in part governed by mitochondria are also commonly deregulated in cancers. Furthermore, patient tumor samples from a variety of cancers have revealed that mitochondrial dynamics machi… Show more

“…Increasing the expression of Drp1 in the neurons of Drosophila under the Ddc-Gal4 transgene seems to decrease longevity, and Drp1 has been found to increase the ROS levels in mitochondria [44]. In our experiments, the lifespan of ies that overexpress Drp1 was signi cantly decreased when compared to controls, which may be due to elevated oxidative stress beyond a threshold of a normal lifespan.…”

“…It is well-known that mitochondrial dynamics are related to the cell death pathways. Mitochondrial dysfunction, including decreased respiration and oxidative phosphorylation (OXPHOS), metabolic shift toward glycolysis, and increased mitochondrial ROS formation, leads to the activation of prosurvival or prodeath pathways, depending on the stimulus (32). We were interested in understanding whether mitochondrial ROS production was involved in cell death induced by ZZW-115.…”

“…Mitochondrial dynamics process can be activated through different signaling pathways; a high content of ROS and the accumulation of damaged mitochondria can activate fission processes in order to decrease oxidative stress and eliminate damaged mitochondria [34,35]. The activation of fission is the first step for mitophagy, the form of autophagy (a process to remove unfolded proteins and clear damaged organelles) that eliminates mitochondria when required [28].…”
