The International Neuroblastoma Risk Group (INRG) Staging System: An INRG Task Force Report

Abstract: A B S T R A C T PurposeThe International Neuroblastoma Risk Group (INRG) classification system was developed to establish a consensus approach for pretreatment risk stratification. Because the International Neuroblastoma Staging System (INSS) is a postsurgical staging system, a new clinical staging system was required for the INRG pretreatment risk classification system. MethodsTo stage patients before any treatment, the INRG Task Force, consisting of neuroblastoma experts from Australia/New Zealand, China, Eu… Show more

“…EFS according to treatment period. High-dose busulfan in high-risk neuroblastoma S Proust-Houdemont et al BM involvement at diagnosis was associated with a worse prognosis, as already demonstrated by Hartmann et al, 32 and a good response to induction therapy seems to be another important prognostic factor in the HR-NBL population, as recently reported by other studies. 1,[34][35][36] It is noteworthy that the criteria required to receive HDC differed from those applied in the HR-NBL1/SIOPEN trial.…”

“…In our neuroblastoma cohort, 69% (27/39) were defined as high-risk disease (defined according to classification systems) (12,13). Among the high-risk patients, 37% (10/27) had mutations in Rho/Rac-associated genes, whereas patients with Rho/Rac gene mutations were found in 25% (3/12) of patients with intermediate-and low-risk neuroblastoma (Table 1).…”

“…The INRG staging system (INRGSS) uses preoperative radiological features and image-defined risk factors (IDRFs) to distinguish low-risk tumors from high-risk tumors [48,49]. The INRG classification system was developed based on analysis of 13 clinical and biological variables for effects on event-free survival rates, including patient age at diagnosis, INRGSS tumor stage, serum lactate dehydrogenase and ferritin levels, tumor histological category, grade of differentiation, tumor [51,52] (Figure 3).…”
