The International Mouse Phenotyping Consortium: comprehensive knockout phenotyping underpinning the study of human disease

Abstract: The International Mouse Phenotyping Consortium (IMPC; https://www.mousephenotype.org/) web portal makes available curated, integrated and analysed knockout mouse phenotyping data generated by the IMPC project consisting of 85M data points and over 95,000 statistically significant phenotype hits mapped to human diseases. The IMPC portal delivers a substantial reference dataset that supports the enrichment of various domain-specific projects and databases, as well as the wider research and clinical community, wh… Show more

“…( A ) Venn diagram illustrating that of 3073 proteins with significant correlation to at least 1 Ca 2+ wave parameter of interest, 647 had orthologues with glycemic SNPs. ( B ) Of the 647 candidates, 9 were previously suggested to be secreted (61-64), 20 had existing KO mice with at least one metabolic phenotype (65), and 36 had existing compounds targeting them (57-60). ( C ) Example comparisons of strain variance for proteins (left column, (4)) and Ca 2+ traits (right column).…”

“…For example, the deep integration between OBA and HPO will facilitate the use of gene-phenotype associations derived from GWAS studies in variant prioritisation software such as Exomiser 75 , which is used for clinical diagnostics. This has the potential to significantly extend the existing sources of gene-phenotype data from annotations of Mendelian disease resources such as OMIM and Orphanet as well as model organism resources such as MGI 5 , IMPC 7 and ZFIN 76 .…”

“…The contribution of individual amino acids such as isoleucine to propionic acid production is unknown and will be addressed. We suggest the study of Acad8 and Acadsb KO mouse models, which are available through the International Mouse Phenotyping Consortium (Groza et al 2022).…”
