Deterioration of the immune system due to antibiotic therapy can be restored by immunomodulator application. In this paper we estimate the effect of ampicillin, amikacin, doxycycline, rifampicin, rifamycine and immunomodulator Tołpa Peat Preparation (TPP) on neovascular reaction induced in murine skin by human mononuclear cells (MNC) injection. MNC originating from 15 healthy volunteers were injected intradermally to Balb/c mice. Antibiotics (3, 15, or 75 mg/kg of body weight) alone or with TPP (10 mg/kg of body weight) were administrated subcutaneously to mice on three consecutive days. The number of newly formed blood vessels was measured in dissection microscope 72 hours after cell injection. Results: TPP stimulated angiogenic activity of MNC at the dose 5 and 10 mg/kg. Rifamycine exerted strong stimulatory action, ampicillin slightly stimulated immune response, while doxycycline and rifampicin downregulated it. Amikacin did not influence the results of angiogenesis tests. Studied antibiotics (15 mg/kg), except rifamycine, inhibit the angiostimulatory effect of the tested immunomodulator. TPP should be applied after antibiotic therapy to maintain its stimulatory effect and restore proper host immune function.

An electrochemical HbA1c sensor with high sensitivity and good specificity is proposed based on the electrochemical immune principle. The reproducibility and conductivity of the electrode are improved by depositing gold nanoparticles (AuNPs) on the surface of the screen-printed electrode (SPE). The HbA1c antibodies are immobilized on the surface of the modified electrode by adsorption to capture the HbA1c in the sample. The hindering effect of HbA1c on the electrode transfer reaction was exploited as the HbA1c detection mechanism. The electrode’s properties were characterized by electrochemical impedance spectroscopy (EIS), and the measurement properties of the electrode were analyzed using differential pulse voltammetry (DPV) and cyclic voltammetry (CV). The experimental results show that the peak current signal of the electrochemical immunosensor produced a linear response to HbA1c in the concentration range of 20–200 μg/mL, a linear relationship coefficient of 0.9812, a detection limit of 15.5 µg/mL, and a sensitivity of 0.0938 µA/µg·mL−1. The sensor delivered satisfactory repeatability, stability, and anti-interference performance. Due to its small size, high sensitivity, and wide linear detection range, it is expected to play a significant role in managing diabetes at home.

RESUMO:Considerando-se a importância do timo para o sistema imunológico e o potencial imunodepressor da gentamicina, este estudo objetivou avaliar os efeitos deste aminoglicosídeo sobre a histologia do timo de coelhos da raça Nova Zelândia Branco. Para isto, utilizou-se 20 coelhos adultos, 10 machos e 10 fêmeas, com peso médio de 2,5 kg, hígidos, da raça Nova Zelândia Branco, os quais foram divididos em dois grupos (controle e tratado) com cinco machos e cinco fêmeas cada. Após sete dias de tratamento diário com 4,4 mg/kg de gentamicina por via subcutânea, no oitavo dia, os animais foram sacrificados e seus timos, após coletados, foram fixados em solução de Bouin por 24 horas e processados, rotineiramente, para a inclusão em parafina. Após a microtomia semi-seriada, à espessura de 5 µm, com intervalos de 1000 µm, os cortes histológicos foram corados pelas técnicas do Tricrômico de Masson. Analisou-se, por meio de microscopia de luz, as características morfológicas da cortical e medular dos lóbulos tímicos individualizados destes cortes. Não foram observadas diferenças histológicas entre os sexos ou entre os grupos controle e tratado. Os resultados demonstraram que este antibiótico, utilizado em dose terapêutica e pelo período recomendado, não alterou a morfologia tímica dos animais tratados.Palavras-chave: Aminoglicosídeo. MORPHOLOGY OF THE THYMUS OF NEW ZEALAND WHITE RABBITS TREATED WITH GENTAMICIN SUMMARY:Considering the thymus importance for the immune system and the immunodepressive potential of gentamicin, this study aimed to evaluate the effects of this aminoglycoside on the thymus histology of New Zealand White rabbits. For this, it were used 20 adult rabbits, 10 males and 10 females with a mean weight of 2.5 kg, healthy, of the New Zealand White breed, which were divided into two groups (control and treated), with five males and five females each. After seven days of daily treatment with 4.4 mg/kg of gentamicin by subcutaneous injection, on the eighth day the animals were sacrificed and their thymuses, after collected, were fixed in Bouin solution for 24 hours and processed routinely for embedded in paraffin. After the semi-serial microtome with thickness of 5 µm at intervals of 1000 µm, the histological sections were stained using Masson's Trichrome techniques. It were analyzed, by light microscopy, the morphological characteristics of cortical and medullary of thymic individualized lobes of these sections. It was not observed histological differences between sexes or between control and treated groups. The results showed that this antibiotic, used in therapeutic dosage and by the recommended period, did not alter the morphology of the thymus of treated animals.
