The interaction of the cell-contact proteins VASP and vinculin is regulated by phosphatidylinositol-4,5-bisphosphate

Abstract: Focal contact assembly involves interaction between VASP and vinculin, which is enhanced by PIP2-induced vinculin activation and oligomerization. Given that vinculin and VASP both bind to F-actin, vinculin-VASP complexes might bundle the distal ends of actin filaments in focal contacts. We propose that PIP2-dependent signalling modulates microfilament organization at cellular adhesion sites by regulating vinculin-VASP complexes.

“…When bound by lipid and/or actin talin can exist as an antiparallel homodimer (46), whereas vinculin has been proposed to form homodimers or trimers after its interaction with vasodilator stimulatory protein (47). In this scenario a vinculin trimer has been proposed to bind to the antiparallel talin homodimer to provoke changes in the actin cytoskeleton (23).…”

“…In Vitro Chemical Cross-linking-Chemical cross-linking of recombinant VT, MVT, and VH was performed essentially as described (13). Purified (M)VT alone with VH or mixtures of both tail domains (100 pmol, 3.5 M) were incubated in the presence or absence of lipids at 37°C for 30 min.…”

“…Anchorage of microfilaments is mediated by VT that harbors two actin binding regions (11,12). Through the recruitment of the vasodilator-stimulated phosphoprotein VASP and the Arp 2/3 complex to the hinge region, vinculin is also tightly linked to actin dynamics at cell adhesion sites (13)(14)(15). Ligand interactions are regulated by an intramolecular association of VH and VT, which masks the binding sites for most ligands (12,(15)(16)(17)(18)(19).…”

“…Phospholipids were prepared as described in Huttelmaier et al (39). Turkey gizzard FLN was incubated without or with a 5-fold molar excess of phosphatidylinositol 4,5-bisphosphate or phosphatidylserine and recombinant FLNa fragments were incubated without or with a 5-fold molar excess of phospholipids (phosphatidylcholine:phosphatidylserine: phosphatidylinositol 4,5-bisphosphate ϭ 9:9:2) for 30 min at 4°C and immobilized on nitrocellulose membrane (5-20 pmol of protein/spot) using a dot blotter with circular slots (Biometra, Göttingen, Germany).…”
