The interaction between EAP30 and ELL is modulated by MCM2

Abstract: Edited by Ivan Sadowski Keywords:Eleven-nineteen lysine-rich leukemia ELL-associated protein 30 Mini-chromosome maintenance 2 a b s t r a c t ELL-associated protein 30 (EAP30) was initially characterized as a component of the Holo-ELL complex, which contains the elongation factor ELL. Both ELL and Holo-ELL stimulate RNA pol II elongation in vitro. However, ELL and not Holo-ELL inhibits RNA pol II initiation. It is not clear how these two discrete functions of ELL are regulated. Here we report that mini-chromos… Show more

“…Similarly, EAP30—a subunit of ESCRT‐II—controls HIV‐1 RNA trafficking and gene expression through a complex formed by HIV‐1 Gag, ESCRT‐II and Staufen‐1 . The mechanism by which EAP30/ESCRT‐II facilitates HIV‐1 genomic RNA trafficking remains unclear, although—considering the roles for ESCRT‐II in the nucleus —EAP30/ESCRT‐II is likely part of the RNP complex that mediates the nuclear export of viral RNA. Other partners of EAP30, such as EAP45, have an RNA‐binding domain that is likely conserved , and RILP—which associates with EAP30—can also have an effect on the localization of viral RNA in the cytoplasm.…”

“…ELL has been found to both positively and negatively regulate RNA polymerase II activity, whereas the holo‐ELL complex (comprising ELL and the three ESCRT‐II components) only stimulates activation (Schmidt et al, 1999). Recently, Vps22 was discovered to interact with the helicase MCM2 (mini‐chromosome maintenance 2), which is involved in DNA replication (Mathews et al, 2009). It seems that ELL and MCM2 competitively interact with Vps22, implying that MCM2 regulates the stability of holo‐ELL and thus at least partly determines whether RNA polymerase II activity is suppressed or stimulated.…”
