The inotropic agent digitoxin strengthens desmosomal adhesion in cardiac myocytes in an ERK1/2-dependent manner

Abstract: Desmosomal proteins including desmoglein‐2 (DSG2), plakoglobin (PG), and desmoplakin (DP) are components of the intercalated disc and mediate cardiac myocyte cell‐cell adhesion. Enhancement of cardiac myocyte cohesion, referred to as “positive adhesiotropy”, was demonstrated to be a function of sympathetic signaling in the heart and to be relevant for a sufficient positive inotropic response to adrenergic stimulation. Here we used the positive inotropic agent digitoxin to further investigate the link between i… Show more

“…Mitochondria are remarkably dynamic organelles that perform diverse yet interconnected functions by producing ATP and multiple biosynthetic intermediates that participate in cellular stress responses, autophagy, and apoptosis ( Ma et al, 2020 ; Pflüger-Müller et al, 2020 ; Sawashita et al, 2020 ). The mitochondrial membrane potential is a key indicator of mitochondrial activity as it reflects the processes of electron transport and oxidative phosphorylation, which enable ATP production ( Schinner et al, 2020 ; Sørensen et al, 2020 ). High-glucose conditions have been shown to disrupt the mitochondrial membrane potential and increase the ATP levels ( Nesti et al, 2020 ).…”

“…The plate was constantly shaken for 10 min to fully dissolve the purple crystals. Then, the OD values were measured in an enzyme-linked immunosorbent detector at 490 nm wavelength [ 32 , 33 ].…”

“…Cells were fixed with 4% paraformaldehyde at room temperature for 1 h, and then were treated with 0.5% Triton X-100 for 1 h. The cells were washed twice with phosphate-buffered saline, and then apoptosis was analyzed with a TUNEL Apoptosis Assay Kit (Beyotime Biotechnology) in strict accordance with the manufacturer’s instructions ( Santosa et al, 2020 ). Finally, the nuclei were stained with 0.5 g/mL 4′,6-diamidino-2-phenylindole for 4 min ( Schinner et al, 2020 ). Apoptotic cells (those with nuclei dyed brown-yellow) were observed and photographed under a fluorescence microscope.…”
