The influence of Ostertagia circumcincta and Trichostrongylus colubriformis infections on the pharmacokinetics of febantel in lambs

Abstract: Plasma concentrations of febantel and its major metabolites fenbendazole, oxfendazole and oxfendazole sulphone were determined after oral administration of 7.5 mg/kg febantel in lambs before and 28 days after infection with 50,000 L3 larvae of Ostertagia circumcincta or Trichostrongylus colubriformis. The febantel concentrations were always very low and only in a few samples higher than the detection limit. The mean decrease in AUC for the three metabolites for the infected sheep in comparison to the parasite … Show more

“…In the present research, we examined the pharmacokinetics of febantel after oral administration to healthy tiger puffer, in order to determine if the drug is also metabolized to fenbendazole in tiger puffer as in other animals (Baert et al, 1993;Debackere et al, 1993;BenZvi et al, 1996), and also if febantel, more specifically fenbendazole, is absorbed into blood of the host. We also report the potentially significant anti-H. okamotoi activity of febantel in both tiger puffer experimentally infected and those naturally infected with this parasite.…”

“…This concentration was at least 10 times lower than those of the febantel metabolites (Debackere et al 1993). However, it seems that it is mainly taken up via the surface, at least when it is given as a single drug.…”

“…Kidney deficiency may hinder drug excretion (Guerrini et al, 1984;Terao & Shen, 1984). Gastrointestinal diseases may decrease the rate and extent of absorption of drugs administered orally (van Gogh et al, 1984;Debackere et al, 1993). Diseases of the heart and circulatory system may influence the distribution of drugs in the body (Benowitz & Meister, 1976;Sweeney et al, 1993).…”
