The inflammasomes, immune guardians at defence barriers

Abstract: SummaryAs a result of its strategic location, the epithelium is constantly exposed to a wide variety of pathogen and danger signals. Traditionally, the epithelium has been perceived as a defensive but passive barrier; however, it has now become evident that the epithelium senses and actively responds to these signals in order to maintain barrier homeostasis and contributes to the inflammatory response. One way it does this is by producing pro‐inflammatory cytokines including interleukin‐1β (IL‐1β) and IL‐18. T… Show more

“…Indeed, IL-1 is constitutively expressed in many epithelial cells, where it helps defend against pathogens and injury ( Figure 1E) (138). However, loss of control can lead to inappropriate activation, chronic inflammation and disease (139).…”

“…Inflammatory bowel disease (IBD) is a chronic inflammatory disorder of the gut that includes ulcerative colitis (UC) and Crohn's disease (CD) [1]. While the incidence of IBD only occurs in 1-2% of all colorectal cancer (CRC) patients in the general population [2], this incidence has been quantified to increase up to 5% in patients suffering over 20 years of the disease [3].…”

“…However, KM1608 did not induce the expression of tumor necrosis factor-α (TNF-α). Different from other cytokines, IL-1β is synthesized from a precursor and requires activation by inflammasome [39]. IL-1β controls the recruitment of neutrophils to the infection site to destroy the damaged tissue [40].…”
