The Increase in Retinal Cells Proliferation Induced by FGF2 is Mediated by Tyrosine and PI3 Kinases

Abstract: Since 1973, multiple effects of basic fibroblast growth factor have been described in a large number of cells. These effects include proliferation, survival and differentiation. The aim of this work was to study the intracellular pathways involved in the basic fibroblast growth factor (FGF2) effect on rat retinal cells proliferation in vitro. Our data show that treatment with FGF2 increases proliferation in a concentration- and time-dependent manner. The effect of 25 ng/ml FGF2 was blocked by 10 microM geniste… Show more

“…PRDM5 is a member of a family of transcriptional regulators that regulate cell growth and differentiation, and which modulate aspects of both canonical and non-canonical Wnt signaling. 67,68 FGF2 is expressed in surface ectoderm overlying the optic vesicle, controls division of the neuro-epithelium into neuronal retina and RPE, 69 and was selected due to these factors as well as its involvement in lens morphogenesis, 70 retinal cellular proliferation, 71 and retinal ganglion cell axonal guidance. 72 However, no diseasecausing variants were identified in PRDM5 and FGF2, or three other candidate genes (CIC, POU2F2, and SOS2).…”

“…Additionally, injections of FGF2 alone following excitotoxic injury did not promote regeneration in rat retinas [143]. Nevertheless, FGF2 has the potential to accelerate the proliferation of Müller cells derived from rat retinas in vitro [144]. Moreover, in vivo studies demonstrate that FGF2 could stimulate retinal regeneration in combination with insulin or Wnt signalling activator CHIR99021 [145,146].…”
