The in vitro effects of dehydroepiandrosterone on human osteoarthritic chondrocytes

Abstract: Our study demonstrates that DHEA has no toxic effect on chondrocytes up to 100 micro M of concentration and has an ability to modulate the imbalance between MMPs and TIMP-1 during OA at the transcription level, which suggest that it has a protective role against articular cartilage loss.

“…MMP-1 is a commonly detected metalloproteinase in synovial fluid from patients suffering from osteoarthritis (Freemont et al , 1997; Goupille et al , 1998; Kanyama et al , 2000; Yoshihara et al , 2000; Neuhold et al , 2001; Jo et al , 2003; Little et al , 2009). Another degradative enzyme, ADAMTS-4, was reported to be a major aggrecanase in cartilage of mouse and ADAMTS-5 was known to be important in cartilage matrix destruction during osteoarthritis (Stanton et al , 2005; Echtermeyer et al , 2009).…”

“…DHEA increased human a1 (I) procollagen and reduced collagenase (MMP-1) gene expression in cultured skin fibroblasts (Lee et al, 2000). In human cultured chondrocytes, DHEA increased type II collagen and TIMP-1 gene expressions, but decreased type I collagen, MMP-1, and MMP-3 expression (Jo et al, 2003). On the other hand, DHEA-S stimulated collagenase and gelatinase production, but did not alter collagen production in rabbit cervical tissue (Sakyo et al, 1987).…”
