Background: Whole plant of Bambusa arundinaceae (Bamboo) belonging to family (Graminae) is used in number of disease including liver ailments in various parts of India. The present study is done to evaluate antioxidant and Hepatoprotective activity of methanolic extract of young shoots of Bambusa arundinaceae in Thioacetamide induced liver injury in rats. Material and method: Antioxidant activity of methanolic extract was evaluated by DPPH, Ferric reducing power assay and Lipid peroxidation. In vitro hepatoprotective activity of methanolic extracts (25, 50, 100, 200 and 400 µg/ml) against thioacetamide (100mM) induced cytotoxicity was assessed by monitoring cell viability in HepG2 and Hep3b tumor cell line and also in primary hepatocytes. In vivo hepatoprotective activity of methanolic extract (50, 100, 200 mg/kg p.o.) was observed againt thioacetamide (100 mg/kg s.c.) induced liver injury in rat by measuring biochemical parameters viz, Aspartate Transaminase, Alanine Transaminase, Alkaline Phosphatase, Total and Direct Bilirubin. The histopathological studies were also performed. Results: Methanolic extract of young shoots showed good antioxidant activity. Thioacetamide caused significant reduction in cell viability in in vitro studies. Treatment with methanolic extract caused significant increase in cell viability. In vivo studies showed that Thioacetamide induced elevation of biochemical parameters in rats were significantly (p<0.0001) decreased with methanolic extract treatment in rats. Histopathology studies also supported the protective effect of methanolic extract. Conclusion: The current study reveals that the methanolic extract of Bambusa arundinaceae has hepatoprotective activity which may be due to its antioxidant activity.

The seasonal variation of CuCl2-mediated low density lipoprotein (LDL) oxidation (10 microM Cu2+, lag phase, rate of oxidation and maximum absorbance at 234 nm) were measured in 43 men and women on 4-6 occasions (mean 5.7 +/- 0.5) over a 12-month period. The lag phase averaged 52.7 +/- 0.6 min and did not differ by gender. Lag phase and rate of the rapid propagation phase of LDL oxidation showed a sinusoidal pattern over the year (increased and reduced oxidative susceptibility during January and June-July, respectively; both p < 0.001). Changes in plasma alpha-tocopherol, ascorbic acid, lycopene or beta-carotene concentrations did not explain seasonal differences in oxidative susceptibility of LDL in vitro. Nor did plasma lipid content of linoleic acid, the main substrate of lipid peroxidation, vary. However, the amount of hydroperoxy- plus hydroxy-fatty acids in plasma lipids varied according to season (p < 0.024) and was related to the lag phase (r = -0.26, p < 0.001). Seasonal variation in oxidative susceptibility was not significant after adjusting for hydroperoxy- plus hydroxy-fatty acids (p = 0.506). Isolated LDL is more vulnerable to Cu2+-induced lipid peroxidation during the winter and this may be due to the higher amount of oxidised lipids during that period.

Clinical and epidemiological studies have provided circumstantial evidence that oxidized low-density lipoprotein (LDL) and antioxidants are involved in the pathogenesis of atherosclerosis. Superoxide dismutases (SODs) have been shown in vitro to protect LDL from deleterious effects of superoxide anions. In the present study, we have used adenoviral gene transfer to determine effect of extracellular SOD (EC-SOD) on atherogenesis in LDL receptor -/- mice. Intravenous administration of EC-SOD adenovirus (2 x 10(9) plaque forming units) into tail vein targeted transgene mainly to liver and induced a 3.5- to sevenfold increase in plasma total SOD activity. EC-SOD was secreted into circulation for 2-3 weeks mostly in a truncated B-form, suggesting that endogenous proteolytic mechanisms control the level and distribution of the enzyme. Therapeutic potential was determined by measuring plasma resistance against copper oxidation and analyzing atherosclerotic lesion areas in aortas of LDL receptor -/- mice. Mice were kept on a cholesterol diet for 10 weeks before gene transfer and 3 or 6 weeks after the gene transfer. Results showed a tendency for a reduction in the overall lesion area after EC-SOD gene transfer as compared with LacZ transduced control mice, but the difference did not reach statistical significance. It is concluded that short-term overexpression of EC-SOD in vivo does not affect atherogenesis in LDL receptor -/- mice.
