Oviductal factors may be obtained by ultrafiltration of conditioned medium, added to a simple media and used in bovine embryo culture. In this study, we aimed to analyze the development of bovine embryos produced with oviductal factors compared to those cultured in the presence of BSA or serum, the effects of glucose in presence of these protein supplements, and the ability of oviductal factors to support embryo development during the entire culture period. In vitro produced bovine zygotes from slaughterhouse ovaries were cultured in modified-synthetic oviduct Ž . Ž . Ž . Ž . fluid mSOF alone or supplemented with 1 oviductal factors, 2 BSA and 3 FCS. Oviductal factors showed embryotrophic activity, although with blastocyst rates lower than those in BSA and Ž . FCS. Glucose 1.5 mM added at Day 2 of culture did not affect development in the presence of oviductal factors. The number of cells in expanded blastocysts was unaffected by the presence of glucose or any of the protein supplements used. Both BSA and FCS, respectively, improved blastocyst rates of Day 6 embryos produced with oviductal factors. The effect of oviductal factors was masked by the presence of BSA during the entire culture. FCS promoted an earlier appearance of blastocysts. It is concluded that the effect of glucose on in vitro embryo development depends upon the source of protein. Oviductal factors are not an appropriate supplement for embryos beyond Day 6 of culture in SOF, although blastocyst rates of such embryos may be increased by culturing them in the presence of FCS or BSA. q

Abstract.To improve the development of singly cultured bovine embryos, we developed a co-culture method with trophoblastic vesicles. The growth of trophoblastic cells was markedly increased in vitamin-supplemented medium 199 compared with medium 199. Upon co-culture of a single embryo with trophoblastic vesicles in vitamin-supplemented medium 199, embryo development to the blastocyst stage was significantly higher than in embryos co-cultured with trophoblastic vesicles in RPMI 1640 or with cumulus cells in medium 199 (control). In the absence of the vitamin cocktail, co-culture with trophoblastic vesicles in medium 199 did not improve embryo development compared with that of the control. The vitamin cocktail was effective in embryo development when co-cultured with trophoblastic vesicles, but not with cumulus cells. Embryo development was not improved in the absence of co-cultured trophoblastic vesicles, even in the presence of vitamin cocktail. In conclusion, the co-culture system with trophoblastic vesicles in vitamin-supplemented medium 199 efficiently enhances the development of singly cultured embryos. Dev. 58: [717][718][719][720][721] 2012) T here has long been the need for a specific culture system for use with a small number of oocytes collected individually from one or both ovaries of a bovine under the guidance of an ultrasonic apparatus. Generally, the development of singly or sparsely cultured early embryos is poor compared with embryos cultured in larger numbers [1,2]. An individually cultured oocyte becomes well developed upon co-culturing with granulosa cells, but blastocyst formation remains low [3]. Fujita et al. [4] suggested that embryo culture medium was effective for development of an individually cultured embryo. Larson et al. [5] reported that interferon-τ was more highly secreted from embryos cultured in groups compared with an individually cultured embryo. It is well known that interferon-τ secreted from trophoblastic cells is an important signal of maternal recognition during pregnancy [6][7][8][9][10]. Interferon-τ mRNA is detected at the blastocyst stage, and interferon-τ is first produced after blastocyst expansion followed by hatching from the zona pellucida [11]. The development of embryos is promoted by recombinant interferon-τ, with its receptor expressed in morula-stage embryos [12]. Embryos effectively develop upon co-culture with trophoblastic vesicles derived from ex vivo cultures [13,14]. Stojkovic et al. [15,16] reported that either interferon-τ secreted from trophoblastic vesicles or trophoblastic vesicle-conditioned medium improves the development of bovine embryos in vitro. This study was conducted to examine the hypothesis that nitric oxide (NO) affects prehatching development of bovine oocytes fertilized in vitro. In experiment 1, inseminated oocytes were cultured in a cumulus–granulosa cell (CG) coculture system to which 0.008 or 0.04 mM of sodium nitroprusside (SNP), a spontaneous NO releaser, was added at 18 or 60 hr postinsemination. Embryo development was greatly (P < 0.001) inhibited by the addition of SNP, regardless of time of addition or SNP concentration. In experiment 2, eight‐cell embryos were cultured singly in a defined medium, to which 0.0016, 0.008, or 0.04 mM of SNP was added. Development to the blastocyst stage was greatly (P < 0.001) decreased after addition of SNP compared with no addition. Higher (P < 0.02) concentration of NO metabolites was found in developmentally arrested embryos than in developing embryos at 144 hr postinsemination (experiment 3). In experiment 4, blastocyst formation of oocytes cocultured with CGs was significantly (P < 0.02) increased after addition of hemoglobin (Hb, 1 μg/ml), an NO scavenger. Prehatching development of oocytes was significantly (P < 0.05) increased after addition of Hb at different time intervals (18, 60, or 144 hr postinsemination) in experiment 5. Embryo development was not enhanced by Hb addition to the culture medium in the absence of CGs (experiment 6). Prehatching development of eight‐cell embryos derived from a Hb‐containing culture system was not promoted by the further addition of Hb after transfer of the embryos to a defined and CG‐free single‐embryo culture system (experiment 7). In conclusion, NO, which may be secreted from CGs, has an inhibitory role in prehatching development of bovine oocytes fertilized in vitro, and use of an NO scavenger, Hb, in a coculture system enhances blastocyst formation.
