The Importance of Adjuvant Formulation in the Development of a Tuberculosis Vaccine

Abstract: An effective protein based vaccine for tuberculosis (TB) will require a safe and effective adjuvant. There are few adjuvants in approved human vaccines, including Alum and the oil-in-water (o/w) based emulsions MF59 (Novartis Vaccines and Diagnostics), AS03 and AS04 (GlaxoSmith Kline Biologics, GSK) AF03 (Sanofi), and liposomes (Crucell). When used with pure, defined proteins, both Alum and emulsion adjuvants are effective at inducing primarily humoral responses. One of the newest adjuvants in approved product… Show more

“…The protection provided by the TIV is strong in young adults, but its efficacy decreases in the elderly due to immunosenescence, which is characterized by decreases in effector cell number and function, as well as alterations in the production of inflammatory and antiviral cytokines (3)(4)(5). Additionally, individual immune responses vary dramatically because of multiple factors, including the immunogen, route of administration, age, and virus type.…”

“…1,2 When formulated with the Toll-like receptor (TLR)-4 ligand synthetic glucopyranosyl lipid adjuvant (GLA), ID93 induces T helper type 1 immune responses in mice, guinea pigs, and non-human primates. 1,3 Phase I clinical trials designed to evaluate the safety and immunogenicity of ID93 with an oil-in-water emulsion formulation containing GLA (GLA-SE) are ongoing. 4,5 Liposomes represent an alternative method of formulation of TLR4 ligands such as GLA.…”

“…The MPL stimulate increased numbers of DCs and monocytes in the draining lymph nodes and promote IFN-γ production by specific CD4+ T cells, therefore, shift the immune response toward a Th1 profile. 28,29 Antibodies provide the first line of defense against infection by microbial pathogens. 30 Our immunogenicity experiments showed that the vaccine formulated in NLX/alum significantly increased total antibodies vs. alum-adjuvanted vaccine group.…”
