Specific microorganisms can produce bacterial nanocellulose (BNC), with acetic acid bacteria (AAB) being the most active producer. The family Acetobacteraceae includes the obligate aerobic, motile acetic acid bacteria. The BNC has attracted a lot of interest across a wide range of industries, including pharmaceuticals, due to its flexible characteristics, properties, and advantages. The present study was conducted to purify and characterize BNC produced from AAB isolated from apple vinegar. Bacterial nanocellulose was synthesized using a natural date palm liquid medium at pH 6 at 30°C for 8–10 days. The bacterial cellulose produced was then purified using a technique involving 0.1 M sodium hydroxide. To ascertain the surface morphology, size, and form of the BNC membrane, three techniques were used for characterization: X-ray diffraction (XRD), atomic force microscopy (AFM), and transmission electron microscopy (TEM). The results of the XRD analysis confirmed that the BNC particle size ranged between approximately 17.10 and 70.33 nm, while the AFM analysis revealed that the mean diameter of these nanofibers was 26.58 nm. The TEM images clearly showed that the diameters of the BNC fibers ranged between approximately 26-66 nm. The findings of this study reveal that the characterization of the purified BNC using the XRD, AFM, and TEM analyses showed the presence of fibers with varying nanoscale diameters.

Nanotechnology refers to the synthesis, characterization, production, and manipulation of these particles. Incorporating or encapsulating them with suitable nanomaterials can improve their pharmacokinetic characteristics and efficacy. 5,6 Medicinal plants form capping layers, which affect the size and structure of nanoparticles. 7,8 The medicinal plant Allium sativum, more often known as garlic, was used in the synthesis of silver nanoparticles (AgNPs). 9,10 The plant has medicinal properties and is used to treat a variety of diseases in most cuisines around the world. This spice is reported to have anticancer, antimicrobial, antidiabetic, hypoglycemic, hypolipidemic, antiemetic, and immunomodulatory activities. 11 Allium sativum contains a lot of is abundant in tannins, saponins, phenols, alkaloids, carotenoids, and flavonoids, according to phytochemical studies, and it has been shown to have significant antioxidant activities. 12 The present study was aimed at biologically synthesizing AgNPs using A. sativum ethanolic extract and evaluating their antibacterial and antioxidant capabilities. The global COVID-19 pandemic was caused by SARS-CoV-2. Human cells ingest this virus when ACE2 identifies it. TMPRSS2 prepares SARS-CoV-2 for entry. The clinical outcomes of COVID-19 are associated with ACE2 and TMPRSS2 gene expression polymorphisms. Objective: To determine if the TMPRSS2 gene rs 2070788 SNP in intron 11-12 is associated with severe COVID-19 in Iraqi patients Methods: The study included 120 COVID-19 patients from three Ramadi City hospitals and 80 healthy controls. DNA extraction was done with Wizard genomic TM DNA Extraction Kit, RNA extraction was done with One Script Plus cDNA Synthesis Kit, and qRT-PCR was used for investigating genetic polymorphism.Results: The TMPRSS2 rs2070788 SNP had three genotypes (CC, CT, and TT) and two alleles. The genotypes of COVID-19 patients and controls were compatible with Hardy-Weinberg equilibrium (HWE). Severe COVID-19 patients and controls had significantly higher TT genotype frequencies in TMPRSS2 (28% vs 4%, OR = 9.33; 95% CI = 2.03 to 43.01; p = 0.002) and T allele (48% vs 16%, OR = 2.37; 95% CI = 1.32 to 4.26; p = 0.005), respectively. TMPRSS2 mRNA expression was much lower in severe cases than controls. In addition, the relative expression of TMPRSS2 mRNA was increased by 1.15 ± 0.71 folds in the CC genotype of rs2070788 SNP compared to the CT (0.632 ± 0.25) and TT (0.552 ± 0.193) genotypes, but the difference was not significant (p > 0.05). Conclusion:Iraqis were highly susceptible to COVID-19 due to the rs2070788 TT genotype and T allele. Severe cases also downregulated TMPRSS2 gene expression.
