The impact of exenatide (a GLP‑1 agonist) on markers of inflammation and oxidative stress in normal human astrocytes subjected to various glycemic conditions

Abstract: GLP-1 agonists such as exenatide and liraglutide are novel drugs for the treatment of diabetes and obesity. While improvements in glycemic control can rely on an incretin effect, the mechanisms behind the loss of weight following therapy have yet to be completely elucidated, and seem to be associated with alterations in eating habits, resulting from changes in cytokines e.g. interleukin 1β (IL-1β) and oxidative signaling in the central nervous system (CNS). Increased levels of IL-1β and reactive oxygen species… Show more

“… 93 GLP-1 receptor agonists or analogs activate GLP-1 to stimulate cell proliferation, which can improve dopaminergic neurotransmission, neuron and synaptic function, disease motor symptoms, and exert an optimal neuroprotective effect. 94 Ghrelin is mainly produced by enteroendocrine cells and promotes the release of the growth hormone. It also participates in a variety of endocrine regulation processes and exerts anti-apoptotic and anti-oxidant effects.…”

“…GLP-1 inhibits secretion of IL-1β by LPS-stimulated cultured rat astrocytes [12], and a GLP-1 mimetic liraglutide inhibits secretion of IL-1β and TNF by cultured rat astrocytes exposed to advanced glycation endproducts (AGEs) [16]. A GLP1R agonist, exenatide, upregulates glutathione peroxidase in human astrocytes enhancing their antioxidant potential [17]. GLP1R agonists can not only inhibit pro-inflammatory activation of murine microglia and induce their production of nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF) [13], but also block the conversion of murine astrocytes to the A1 phenotype, which is induced by microglia stimulated with α-synuclein fibrils [14].…”

“…The astrocytes, that represent an important central nervous system cell population, have been proposed to contribute to the transduction of anorectic GLP-1R-dependent signals in rats [6]. GLP-1R expression in astrocytes has been described within rat brain [6][7][8] and in a human astrocyte cell line [9]. On the other hand, no colocalization of GLP-1R with the astrocytic marker glial fibrillary acidic protein (GFAP) was observed in any area of the mouse brain examined, including the hypothalamus [10].…”
