The immunoglobulin IGHD gene locus in C57BL/6 mice

Abstract: Four immunoglobulin heavy chain diversity (IGHD) gene subgroups (DFL16, DSP2, DQ52, and DST4) have been identified previously in BALB/c mice. Although the locations of most IGHD genes have been established based on restriction map and Southern blot analysis, a complete mouse IGHD gene locus map at the sequence level is still not available. In addition, a previous restriction fragment length polymorphism study suggested that significant difference in the IGHD gene locus exists between C57BL/6 and BALB/c mice. T… Show more

“…1A). Nucleotide Identity X analysis shows that it contains one previously reported D H gene (DST4.2), which has never undergone D to J recombination (27), and Myef2rpg (myelin basic expression factor 2 repressor pseudogene) (GenBank TM accession number XM_621300). Most notably, the V-D region encodes two genes, Adam6a and Adam6b (a distintegrin and metalloproteinase domain 6a and -b) (31).…”

“…The sequence of the mouse region was established through bacterial artificial chromosome assembly using Sequencher (Gene Codes): RP23-109B20, RP24-275L15, RP23-404D8, and RP23-270B12 (the last two cover the Igh D region) (27). These bacterial artificial chromosomes provide at least 2-fold coverage except in four regions of 12,735, 1087, 528, and 430 bp, which have single strand coverage.…”

“…These V H gene families are partially interspersed with one another but, depending on position, can be divided into proximal (3′ part of the V H cluster, close to IgH-D region, for example, V H 7183), intermediate (e.g., V H S107), and distal (5′ part of the V H cluster, distant from IgH-D region, for example, V H J558) families. 3′ of the V H elements, separated by ~90 kb, lie 10-15 D segments (Retter et al, 2007;Ye, 2004) followed by 4 J H elements. Because to the uniform transcriptional orientation of V, D, and J segments, V(D)J recombination events at the IgH locus result in deletion of the intervening sequence.…”
