TheWacGene Product of Bacteriophage T4 Contains Coiled-Coil Structural Patterns

Abstract: The bacteriophage T4 late gene wac (whisker antigen control) encodes the protein which forms the fibrous structure on the neck of the virion called whiskers. Amino acid sequence analysis of wac gene product, as deduced from the nucleotide sequence, indicate ten alpha-helical domains (19-40 residues long) with coiled-coil structural patterns. These regions comprise about 70% of the entire 486 amino acid sequence. The alpha-helices are separated by short stretches of polypeptide chain which are similar to the lo… Show more

“…The heptad positions shown in the T4 gpwac sequence are those proposed by Sobolev and Mesyanzhinov (33) with the corrections of Tao et al (36). The loops of the gpwac of bacteriophage T4 (33,36) distant (Ͻ80% amino acid identity) T4-type phages had unrelated C-terminal domains (Fig. 3).…”

“…The sequence of the gpwac polypeptide chain of 487 residues is composed of heptad repeats (a b c d e f g) n , with the a and d positions preferentially occupied by apo lar residues and the e and g positions often occupied by charged residues [92]. Such a motif, a characteristic fea ture of α helical coiled coils proteins, is widely distrib uted [93].…”

“…5 Sequence analysis of fibritin reveals a major region (, 80% of the entire sequence) which contains the heptad repeat pattern (abcdefg) n , where a and d are preferably occupied by apolar residues. 10,11 Such a motif is characteristic of an a-helical coiled-coil structure. 12 In fibritin the heptad pattern is interrupted in several places, which suggests the existence of 13 coiled-coil "segments" connected by 12 linker regions ( Figure 2).…”
