The Synechococcus Strain PCC 7942 glnN Product (Glutamine Synthetase III) Helps Recovery from Prolonged Nitrogen Chlorosis

Abstract: We report the cloning and sequencing of the glnN gene encoding a class III glutamine synthetase from the cyanobacterium Synechococcus strain PCC 7942. Mapping of the transcriptional start site revealed a DNA sequence in the promoter region that resembles an imperfect NtcA binding motif. Expression of glnN is impaired in NtcA-and P II -deficient mutants. The only parameter which was negatively affected in the glnN mutant compared to the wild type was the recovery rate of prolonged nitrogen-starved cells with lo… Show more

“…Synthesis of GlnN following nitrogen deprivation is impaired not only in an NtcA-deficient mutant but also in a mutant deficient in P II (Sauer et al, 2000), suggesting a link between P II and NtcA under conditions of nitrogen deprivation. To investigate this potential relationship, protein synthesis patterns of Synechococcus wild-type, P II -and NtcA-deficient mutants were compared (Aldehni et al, 2003).…”

“…One of the genes exhibiting a putative non-canonical NtcA site is glnN, encoding an alternative glutamine synthetase of type III (Reyes et al, 1997). The glnN product helps Synechococcus cells recover more rapidly after a prolonged period of nitrogen chlorosis, although this gene product is not essential for survival (Sauer et al, 2000). Differential affinity of NtcA towards various DNA-binding sites provides a simple mechanism to modulate gene expression in response to a gradual limitation of the nitrogen source.…”

“…In addition to this orthodox promoter structure, which matches that of the Escherichia coli class II Crp-dependent promoters, NtcA can activate from positions further upstream or from sequences not matching the reported consensus (21). NtcA binding motifs were found in a few genes or operons experimentally characterized in S. elongatus (22)(23)(24) and in others predicted to be controlled by NtcA in the cyanobacterium S. elongatus PCC 6301 (25), which is almost identical to S. elongatus. Remarkably, NtcA binding motifs were found in association with 29 of the 30 transcription units in class 4 ( Fig.…”

“…Interestingly, GSIII is also induced in the cyanobacterium Synechococcus during the early stages of nitrogen deprivation (Sauer et al, 2000). In addition, Mock et al (2008) measured increased transcript levels of two NAD(P)H-GOGAT genes (ProtIDs 29861 and 269160) along with a ferredoxin-dependent form of this enzyme (ProtID 269900).…”
