The ‘Galleria bait method’ for detection of entomopathogenic fungi in soil

Abstract: The ‘Galleria bait method’ originally described for trapping of entomoparasitic nematodes in soil has been used successfully also for the detection of naturally occurring entomopathogenic fungi. In many soil samples several species, such as Beauveria bassiana, Metarrhizium anisopliae or Paecilomyces fumosoroseus, could be detected. This method can be applied for various ecological studies independent of the season and the occurrence of soil inhabiting host insects. Zusammenfassung Die “Galleria‐Köder‐Methode” … Show more

“…T. infestans proved to be highly effective as in vivo bait for B. bassiana and M. anisopliae. Satisfactory results with insect bait methods using other insects such as Galleria mellonella L. (Zimmermann 1986, Tarasco et al 1997 and species of Tenebrio L. (Vänninen et al 1989) were reported by other authors. Fungal detection in the present study did not depend on the method used.…”

“…EPF can be isolated either from the insect cadavers during natural epizootics or directly from soil from crop rhizosphere. The two most commonly used methods by researchers for soil isolation are either baiting the soil with an insect host, for example, Galleria mellonella (Zimmermann 1986) or using specific selective media (Veen and Ferron 1966; Doberski and Tribe 1980; Beilharz et al 1982). Since EPF are generally slow growers, other fast growing opportunistic fungi and bacteria dominate while culturing, which is addressed by the selective use of bacterial antibiotics and fungicides.…”

“…The soil collected were screened for presence of fungal pathogens using the Tenebrio molitor L. bait method (Zimmermann 1986). Five hundred grams of each soil samples collected from the four sites were transferred to plastic bowls (10 cm diameter and 20 cm depth), and moistened by spraying with 150 ml of water.…”
