TheErwinia chrysanthemi3937 PhoQ Sensor Kinase Regulates Several Virulence Determinants

Abstract: The PhoPQ two-component system regulates virulence factors in Erwinia chrysanthemi, a pectinolytic enterobacterium that causes soft rot in several plant species. We characterized the effect of a mutation in phoQ, the gene encoding the sensor kinase PhoQ of the PhoPQ two-component regulatory system, on the global transcriptional profile of E. chrysanthemi using cDNA microarrays and further confirmed our results by quantitative reverse transcription-PCR analysis. Our results indicate that a mutation in phoQ affe… Show more

“…For each array, two total RNA samples (3 g each) were indirectly labeled by reverse transcription using an 11-mer oligonucleotide mixture in the presence of amino-allyl dUTP, as described (30). Labeled cDNA samples were then coupled with Cy3 or Cy5 dye esters (GE Healthcare) and purified (30). Cy-labeled wild type and mutant cDNAs were co-hybridized to Agilent Technologies, Inc., custom ECA_11K Genome Arrays according to the manufacturer's instructions and according to Ref.…”

“…Cy-labeled wild type and mutant cDNAs were co-hybridized to Agilent Technologies, Inc., custom ECA_11K Genome Arrays according to the manufacturer's instructions and according to Ref. 30. The probes (60-mers) include all 4521 predicted protein coding genes and 23 Lucidea Universal ScoreCard Controls (GE Healthcare), with each probe printed twice.…”

“…Indeed, at least six of the genes present in this cluster have been described as being induced and/or implicated in responses to various stress conditions. They include trmD, encoding a tRNA (guanine-n1) methyltransferase (55); rpsU, encoding the 30S ribosomal protein s21 (35); cspA, encoding a conserved transcription and translation enhancer (39); and acnA, encoding an aconitate hydratase (26,56). Perhaps most interesting in this cluster is ompR, encoding the response regulator from the EnvZ/OmpR two-component system, identi- Table S2 in the supplemental material, and the conservation score is given in Table S3 in the supplemental material.…”

“…The nine genes selected for analysis encode proteins involved in different metabolic activities. rplI and rplQ (30), glnA, rho, recA, gyrA, gyrB, and pro1A (54), and 16S rRNA are housekeeping genes that have been used as reference genes in similar studies of various bacteria (51,55). Of the nine genes tested, only the 16S rRNA gene was stably expressed at the two temperatures.…”
