The BCSC-1 locus at chromosome 11q23-q24 is a candidate tumor suppressor gene

Abstract: Frequent allelic loss at human chromosome 11q23-q24 occurs in a wide variety of cancers, suggesting that this region may harbor a tumor suppressor gene. By constructing a physical map of the LOH11CR2 minimal region of loss on 11q23-q24 associated with lung and breast carcinomas, we were able to clone a hereditary translocation, t(11;12)(q23;q24), in a patient with early-onset breast cancer and family history of cancer. The breakpoint was found within 6 kb of the BCSC-1 candidate tumor suppressor gene located i… Show more

“…However, comparable deletion frequency of LOH11CR2A was observed for carcinomas of breast (44%, 7/16) and ovary (38%, 5/13). 18 In between D1 and D2 regions, there are two more overlapping candidate genes ROBO3 and ROBO4 (Fig. 1a).…”

“…Low expression of LOH11CR2A transcripts by Northern blot analysis was found in breast, ovarian and lung cancer cell lines. 18 On the contrary, high expression of this gene was found in CNE-2L2 human nasopharyngeal carcinoma cells by microarray analysis. 44 Ectopic expression of Loh11cr2a enhanced cellular aggregation and increased the expression of E-cadherin/a-catenin, 44 thus suggesting its tumor suppressive activity.…”

“…This tumor suppressor is mutated in a large fraction of various human tumors (18), but its sequence has not revealed anything about how it might work to control cell cycle progression or cell growth. Overexpression of BCSC-1 in oocytes by mRNA injection with or without mutation of conserved consensus PKA phosphorylation sites has not shown a consistent effect on progesteroneinduced oocyte maturation to date, which might be expected for a tumor suppressor already expressed at high levels in the cell.…”
