The hypotriglyceridemic effect of biotin supplementation involves increased levels of cGMP and AMPK activation

Abstract: In addition to its role as a carboxylase cofactor, biotin modifies gene expression and has manifold effects on systemic processes. Several studies have shown that biotin supplementation reduces hypertriglyceridemia. We have previously reported that this effect is related to decreased expression of lipogenic genes. In the present work, we analyzed signaling pathways and posttranscriptional mechanisms involved in the hypotriglyceridemic effects of biotin. Male BALB/cAnN Hsd mice were fed a control or a biotin-su… Show more

“…Among water-soluble vitamins, dietary contents of four vitamins (pantothenic acid, biotin, vitamin B2 and B12) were increased by egg yolk supplementation by 5.40 mg/kg diet, 81.1 mg/kg diet, 600 mg/kg diet and 3.74 mg/kg diet, respectively. However, these increases in water-soluble vitamins were smaller compared with the effective dose on diseases in other reports (42)(43)(44). These estimations indicated that both composition of amino acids and water-soluble vitamins in egg yolk are insufficient for prevention of fatty liver induced by the low-protein diet.…”

“…These discrepancies may be explained by the fact that several studies have found that the abundance of selected proteins was not paralleled by the abundance of their mRNAs [7, 43]. In addition, pyruvate carboxylase apoenzyme expression is regulated by biotin at the posttranscriptional level, thereby influencing both protein synthesis and degradation [44].…”

“…In recent years it has been considered that supplementation with pharmacological doses of biotin also leads to endocrine, systemic, and metabolic actions [24,[40][41]. To our knowledge, there are no reports exploring the effect of biotin on vastus lateralis muscle carboxylases.…”

“…In this tissue, there is truly a constant input and output of tricarboxylic acid cycle intermediaries, which accumulate in metabolic conditions such as diabetes. The lower activity of PC in supplemented mice can explain the hypoglycemic effect that the pharmacological treatment of biotin has in diabetic patients [41,42].…”
