The Hormonal Control of Pyruvate Kinase Activity and of Gluconeogenesis in Isolated Hepatocytes

Abstract: Treatment of isolated rat hepatocytes with saturating concentrations of glucagon caused several modifications in the kinetic properties of pyruvate kinase (ATP:pyruvate 2-O-phosphotransferase, EC 2.7.1.40): SO.5 (substrate concentration at half maximum velocity) for phosphoenolpyruvate was about doubled, whereas Vm.., was not changed; the activity measured at 0.15 mM phosphoenolpyruvate (physiological concentration) was reduced 65-80%; and there was also an increase in the Hill coefficient and in the affinity … Show more

“…The main interest of this mechanism is to be under the control of several hormones such as glucagon and insulin which are involved in the control of gluconeogenesis [4]. It superimposes harmoniously on the control by allosteric effectors, since the metabolites that favour the active conformation of the enzyme are also those that prevent its inactivation by phosphorylation and conversely.…”

“…The activity of pyruvate kinase was measured as described previously, at 0.1 5 mM phosphoenolpyruvate [4]. This activity, referred as 2'0 15, represents 45 9', of V , measured at 5 mM phosphoenolpyruvate, when the enzyme is in its active form as obtained upon incubation of a cellular extract for 30 min at 25 "C in the prescnce of 5 mM MgC12.…”
