The Histidine Triad Protein Hint Is Not Required for Murine Development or Cdk7 Function

Abstract: The histidine triad (HIT) protein Hint has been found to associate with mammalian Cdk7, as well as to interact both physically and genetically with the budding yeast Cdk7 homologue Kin28. To study the function of Hint and to explore its possible role in modulating Cdk7 activity in vivo, we have characterized the expression pattern of murine Hint and generated Hint-deficient (Hint ؊/؊ ) mice. Hint was widely expressed during mouse development, with pronounced expression in several neuronal ganglia, epithelia, h… Show more

“…This indicates that the tumor-suppressive effects of Hint1 are not confined to susceptibility to chemical carcinogenesis. Other investigators did not find an increase in spontaneous tumor in Hint1 À/À mice (Korsisaari et al, 2003). This may be because they followed the mice for only 18 months, the number of mice included in their study was not sufficient, and/or their mice have a different genetic background.…”

“…An interaction between human Hint1 and Cdk7 was observed in a yeast-two hybrid assay (38). However, studies with transgenic mice, in which the hint1 gene was deleted, failed to reveal a phenotype consistent with the regulation of Cdk7 (7). On the other hand, evidence from S. cerevisiae indicates that Hnt1 functions as a positive regulator of the Cdk7-homologous Kin28 kinase (3).…”

“…HINT1 appears to be structurally conserved from yeast through mammals suggesting a highly conserved function. However, no grossly observable deficiencies were noted in hint Ϫ/Ϫ mice, suggesting that other redundant genes can functionally compensate (20). HINT enzyme homologues hydrolyze adenosine-5Ј-monophosphoramidate (AMPNH 2 ) to AMP plus the leaving NH 2 group (19,21).…”
