The Hippo Pathway: A Master Regulatory Network Important in Cancer

Abstract: The Hippo pathway is pervasively activated and has been well recognized to play critical roles in human cancer. The deregulation of Hippo signaling involved in cancer development, progression, and resistance to cancer treatment have been confirmed in several human cancers. Its biological significance and deregulation in cancer have drawn increasing interest in the past few years. A fundamental understanding of the complexity of the Hippo pathway in cancer is crucial for improving future clinical interventions … Show more

“…The regulatory module of the Hippo signaling pathway consists of kinases, including sterile 20 like kinase 1 and 2 (MST1/ MST2), Large Tumor Suppressor 1 and 2 (LATS1/ LATS2), Salvador homolog 1 (SAV1) and MOB Kinase activator 1A and 1B (MOB1A/ MOB1B) 58 , 59 . The switching “ON” and “OFF” of the Hippo signaling cascade is largely controlled by this series of core kinases as illustrated in Figure 1 .…”

“…Similarly, differentially CEGs ( Figure 3 ) which are uniquely co-expressed in CSCs but not in GTCs could be indicative of the AML CSC-specific pathways. In this regard, Tyrosine metabolism ( Sun et al., 2020 ), Hippo signalling pathway ( Liu et al., 2021b ), and the Proteoglycans in cancer ( Ahrens et al., 2020 ) are amongst the currently known pathways to be associated with CSC functioning. Accordingly, the pathways and biological processes enriched by differentially CEGs could be further focused on in future studies concerning AML CSC functioning.…”
