The HIF‑1α/miR‑224‑3p/ATG5 axis affects cell mobility and chemosensitivity by regulating hypoxia‑induced protective autophagy in glioblastoma and astrocytoma

Abstract: Human glioblastoma is a malignant and aggressive primary human brain solid tumor characterized by severe hypoxia. Hypoxia can induce autophagy, which may result in chemoresistance and malignant progression of cancer cells. MicroRNAs (miRNAs) have been reported to modulate hypoxia-induced autophagy in various types of cancers. In the present study, we observed that hypoxia-inducible factor (HIF)-1α expression was increased while miR-224-3p expression was decreased under hypoxia in a time-dependent manner in gli… Show more

“…ATG5 can be targeted by multiple miRNAs, including miR-137-3p, miR-142-3p, and miR-224-3p. The ability of these miRNAs to restrain autophagy contributes to the sensitization of cancer cells to anti-cancer agents [95,97,100]. In addition to ATG5, the inhibitory effects of miR-142-3p on therapeutic resistance can be mediated by targeting ATG16L1 [97].…”

“…In addition, the expression of various miRNAs, including miR-33a [69], miR-494 [55], miR-145 [56], miR-191 [57], miR-27a [58], miR-424 [59], miR-205 [70], miR-21 [60], miR-185 [61], miR-101 [71], miR-210-3p [62], miR-224-3p [72], miR-15a [73], miR-21 [63], and miR-107 [64], has been proven to be HIF-1α-dependent in the progression of various cancers. These findings suggest that the HIF-1α-induced oncogenic effect is caused by transcriptional activation of oncogenic HRMs and inhibition of tumor-suppressive miRNAs to some extent.…”

“…Accordingly, sorafenib combination with different compounds, such as EF24 or genistein, overcomes hypoxia-mediated sorafenib resistance in HCC through HIF-1α downregulation [20,48]. HIF-1α silencing is also effective to decrease invasive potential and rise chemosensitivity of human glioblastoma (LN229) and astrocytoma (U-251MG) cell lines to temozolomide [49], and to potentiate sorafenib-mediated apoptosis in HCC cells [50]. Furthermore, microRNA overexpression stimulates apoptotic cell death by inhibiting HIF-1α both in vitro and in vivo HCC models [33,51], among other tumors [52].…”
