The GNB3 C825T polymorphism and essential hypertension: a meta-analysis of 34 studies including 14 094 cases and 17 760 controls

Abstract: The findings are in agreement with a recently proposed causal model for systolic blood pressure, which correlates it with the T allele and the absolute latitude. Further studies are needed in order to fully address questions about the aetiological mechanism of the particular association, as well as to study the effect in populations of African descent.

“…In recent years many studies have reported a correlation between GNB3-C825T and EH in Caucasian, African, South American and Asian populations. Our analysis showed that the GNB3-C825T allele was associated with EH in the overall population, which is in agreement with the results of a previous multi-ethnic population meta-analysis (Bagos et al, 2007). These authors reported that TT versus CC + CT: OR=1.08, 95% CI: 1.01-1.15, P<0.001 and TT + CT versus CC: OR=1.17, 95% CI: 1.06-1.29, P<0.001.…”

“…The 825T allele is associated with alternative splicing of the gene and formation of a truncated but functionally active β 3 subunit and with enhanced G protein activation [18] . This polymorphism has been associated with multigenic disorders [19] , including hypertension [1] , and it seems to be a candidate for explaining human variability in exercise phenotypes. The GNB3 C825T polymorphism plays a minor role in heart rate and body fatness regulation in blacks, as well as in responsiveness of resting …”

“…Searches for potential studies were carried out using keywords described in detail elsewhere [2] as well as personal contact with experts in the field. The following databases (from inception through 01 April, 2011) were screened: Pubmed, Web of Science, EMBASE, Biological Abstracts, LILACS and Google Scholar.…”

“…Polymorphisms in the GNB3 (chromosome 12p) have been associated with essential hypertension (EH) and obesity [2,3], but there is evidence for statistical heterogeneity (i.e. variation in effect estimates beyond chance) [2].…”
