The Glypican proteoglycans show intrinsic interactions with Wnt-3a in human prostate cancer cells that are not always associated with cascade activation

Abstract: Background Prostate cancer occurs through multiple steps until advanced metastasis. Signaling pathways studies can result in the identification of targets to interrupt cancer progression. Glypicans are cell surface proteoglycans linked to the membrane through glycosylphosphatidylinositol. Their interaction with specific ligands has been reported to trigger diverse signaling, including Wnt. In this study, prostate cancer cell lines PC-3, DU-145, and LNCaP were compared to normal prostate RWPE-1 … Show more

“…These HSPGs might be candidates for core proteins of 3-O-sulfated HS catalyzed by HS3ST1. Among them, syndecan-1, glypican-1 and perlecan have been well studied in hormone-independent prostate cancer cell lines [56][57][58] . Syndecan-1 is also associated with the progression and chemotherapy resistance of CRPC tumors in patients 59,60 .…”

“…These Fz receptors utilize several coreceptors, including protein tyrosine kinase 7 (PTK7), 31 muscle-skeletal receptor tyrosine kinase (MUSK), 32 tyrosine kinase-like orphan receptor (ROR1/ROR2), 33 tyrosine kinase related receptor (RYK), 34 syndecan, 35 , 36 and glypican. 37 , 38 However, the Wnt/PCP pathway also uses Celsr1 and Vangl2 receptors, although the ligand-receptor binding interaction is still relatively unknown 39 ( Fig. 2 ).…”
