The Genome-Wide Expression Profile of Saussurea lappa Extract on House Dust Mite-Induced Atopic Dermatitis in Nc/Nga Mice

Abstract: Saussurea lappa has been reported to possess anti-atopic properties. In this study, we have confirmed the S. lappa’s anti-atopic properties in Nc/Nga mice and investigated the candidate gene related with its properties using microarray. We determined the target gene using real time PCR in in vitro experiment. S. lappa showed the significant reduction in atoptic dermatitis (AD) score and immunoglobulin E compared with the AD induced Nc/Nga mice. In the results of microarray using back skin obtained from animals… Show more

“…The validation of murine models in AD studies is highly needed as no standardized model is currently existing which mimics the highly diverse pathomechanisms (inside-out vs. outside-in theory) [215]. Current AD models include cytokine overexpression (e.g., IL-4 [216] and IL-31), genetic mutations leading to dysfunctional protein expression (e.g., Filaggrin f t /flaky tail mice), NC/Nga model [76,217], PAR2 overexpression and house dust mite (HDM) application [215] and Aspergillus fumigatus-induced AD [200,218]. Formerly used in AD research, the topical application of SDS rather induces characteristics of contact dermatitis [215].…”

“…Cytokine-cytokine receptor interaction is associated with the progression of skin-related diseases by regulating the proliferation of skin-derived precursors (SKPs) and SKP differentiation [43]. In addition, cytokine-cytokine receptor interaction has previously been demonstrated to play an important role in the progression of AE [44]. Thus, we inferred that cytokine-cytokine receptor interaction might be closely correlated with the AE progression.…”

“…Moreover, when high levels of TMTH (10 mg/kg) were administered, the serum IgE level was similar to that of the positive control prednisolone. In the current study, prednisolone was administered at a dose of 3 mg/kg, considering similar studies carried out with prednisolone [ 32 , 33 , 34 , 35 , 36 ]. Thus, TMTH could be effectively utilized as a potential remedy for AD management.…”
