The genome sequence of the two-spot ladybird, Adalia bipunctata (Linnaeus, 1758)

Abstract: We present a genome assembly from an individual male Adalia bipunctata (the two-spot ladybird; Arthropoda; Insecta; Coleoptera; Coccinellidae). The genome sequence is 475 megabases in span. Most of the assembly (94.87%) is scaffolded into 11 chromosomal pseudomolecules, with the X and Y sex chromosomes assembled. The complete mitochondrial genome was also assembled and is 21.2 kilobases in length. Gene annotation of this assembly in Ensembl identified 13,611 protein coding genes.

“…Several other ladybird genome assemblies have been published to date, including Henosepilachna vigintioctomaculata ( Zhu et al , 2023 ), Coccinella septempunctata ( Crowley, 2021 ), Propylea japonica ( Zhang et al , 2020 ), Harmonia axyridis ( Ando et al , 2018 ; Boyes & Crowley, 2021 ; Chen et al , 2021 ; Gautier et al , 2018 ), Adalia bipunctata ( Goate, 2022 ) and Cryptolaemus montrouzieri ( Li et al , 2021 ). Here we present a chromosomally complete genome sequence for Halyzia sedecimguttata , sequenced as part of the Darwin Tree of Life Project, which will also contribute to the growing set of genomic resources for Coccinellidae.…”

“…We selected genomes of 11 ladybird beetle (Coccinellidae) species ( Adalia bipunctata [ Wellcome Sanger Institute Tree of Life Programme et al 2022 ], Coccinella septempunctata [ Crowley et al 2021 ], Cryptolaemus montrouzieri [ Li et al 2021a ], Cynegetis impunctata [our unpublished data, NCBI accession: GCA_030704885.1], Halyzia sedecimguttata [ Crowley et al 2023 ], Harmonia axyridis [ Boyes et al 2021 ], Henosepilachna vigintioctomaculata [ Zhu et al 2023 ], Henosepilachna vigintioctopunctata [our unpublished data, NCBI accession: GCA_030704895.1], M. discolor [our unpublished data, NCBI accession: GCA_030674115.1], Novius pumilus [ Tang et al 2022 ], and Propylea japonica [ Zhang et al 2020 ]) and one Coccinelloidea outgroup Dastarcus helophoroides ( Zhang et al 2023 ), as reference species ( supplementary table S1, Supplementary Material online). The genomes were downloaded from the NCBI Genome database.…”

“…Chromosome-scale scaffolds confirmed by the Hi-C data are named in order of size ( Figure 5 ; Table 2 ). The assignment of sex chromosomes was guided by coverage statistics and synteny to Adalia bipunctata (GCA_910592335.1) ( Wellcome Sanger Institute Tree of Life programme et al , 2022 ). While not fully phased, the assembly deposited is of one haplotype.…”

“…Ten-spot ladybirds are close relatives of the two-spot ladybirds, Adalia bipunctata ( Wellcome Sanger Institute Tree of Life programme et al , 2022 ), and occupy similar habitats with both particularly favouring deciduous trees and hedgerows, although the ten-spot is perhaps more arboreal. The harlequin ladybird, Harmonia axyridis ( Boyes et al , 2021 ), an invasive non-native species in the UK ( Roy et al , 2016 ), also favours these habitats.…”
