The genome of the Mediterranean fruitflyceratitis capitata: Localization of molecular markers by in situ hybridization to salivary gland polytene chromosomes

Abstract: We hybridized cloned DNA segments to salivary gland polytene chromosomes of the medfly, Ceratitis capitata, and thus established molecular markers for 24 sites on 6 out of 10 autosomal arms. An additional marker identified a medfly repetitive element that hybridizes to approximately 100 autosomal sites as well as a granular network that is thought to represent the X chromosome. Some of the markers correspond to 9 characterized transcription units, while 17 remain anonymous; at least 3 of the latter are restric… Show more

“…Thus, it seems that D. melanogaster and C. capitata show similar gene organizations for these loci. This conclusion is strengthened by data presented by Zacharopoulou et al (1991), who showed the similar gene organization between the X chromosome ofD. melanogaster and the 5L chromosome of C. capitata.…”

“…Prior to fl uorochrome staining or in situ hybridization, a denaturing step was performed on the chromosome preparations, as described for other fruit fl y analyses (Zacharopoulou et al, 1992;Procunier and Smith, 1993), with slight modifi cations. Briefl y, the slides were incubated in 2 !…”

“…Detection of hybridization signals was performed using the Fluorescent Antibody Enhancer Set for DIG Detection (Roche Diagnostics GmbH). Larval salivary gland polytene chromosome squashes were prepared as described in Zacharopoulou et al (1992). The salivary gland polytene maps of Zacharopoulou (1990) and their revised version of Gariou-Papalexiou et al (2002) were used for mapping the hybridization signals.…”

“…It should be pointed out that medfly autosomes are numbered from 2 to 6 in descending order of their size (Zacharopoulou 1990). Figure 2 summarizes the integrated cytogenetic and genetic linkage maps of microsatellite markers, along with all previously described markers and genes (Zacharopoulou et al 1992;Gariou-Papalexiou et al 2002;Stratikopoulos et al 2008). …”
