The Genetics of Ficolins

Abstract: Ficolins constitute a family of proteins whose biological role has been an enigma for many years. Over the past few years it has become evident that ficolins are part of the innate immune system and function as recognition molecules in the complement system. The 3 human ficolins, ficolin-1 (M-ficolin), ficolin-2 (L-ficolin) and ficolin-3 (H-ficolin or Hakata antigen) are encoded by the FCN1, FCN2 and FCN3 genes, respectively. Phylogenetic studies suggest that ficolins are of ancient origin. Ficolin-3 seems to … Show more

“…We found variabilities in ficolin-2 binding among the different donors (Fig. 1A), as reported previously (17,18). Interestingly though, we also found striking variabilities in ficolin-2 binding within blood samples from each donor collected in the different tube types.…”

“…A patient who was homozygous for this mutation had recurrent infections (Munthe-Fog et al, 2009). Along with MASP1 (MASP-1/3 and MAp44 proteins) and MASP2 (MASP-2 and MAp19 proteins), a number of SNPs have been described for FCNs (Garred et al, 2010;Degn et al, 2011). However, the importance of ficolins in general and the genetic variations in FCN genes in particular remain largely unknown.…”

“…FCN2 and FCN3 contain eight exons, whereas FCN1 has an additional exon in the collagen-like domain encoding four extra Gly-Xaa-Yaa repeats. 2 The ficolin polypeptide chains consist of 299-326 amino acids. Ficolin-1 and -2 are highly homologous molecules (80%), whereas the degree of homology with ficolin-3 is lower (48%).…”
