The genetic variability, phylogeny and functional significance of E6, E7 and LCR in human papillomavirus type 52 isolates in Sichuan, China

Abstract: Background Variations in human papillomavirus (HPV) E6 and E7 have been shown to be closely related to the persistence of the virus and the occurrence and development of cervical cancer. Long control region (LCR) of HPV has been shown multiple functions on regulating viral transcription. In recent years, there have been reports on E6/E7/LCR of HPV-16 and HPV-58, but there are few studies on HPV-52, especially for LCR. In this study, we focused on gene polymorphism of the HPV-52 E6/E7/LCR sequen… Show more

“…No positive selection sites for HPV16 E6 and E7 genes have been reported in central China [ 37 ]. No positive selection sites for HPV52 E6 and E7 genes have been reported in central and Southwest China [ 38 , 39 ]. In addition, selective pressure analysis from non-Asian population showed different results, with 10R, 14H, 83V in HPV E6 and 85G in HPV16 E7 under positive selective pressure [ 40 , 41 ].…”

“…Thus, more studies are needed to prove its role in CC. It was interesting to us that the long control region ( LCR) of HPV16 and HPV52 was found to have a high permanent binding site for the SOX9 transcription factor ( Xi et al, 2017 ; Song et al, 2021 ; Mane et al, 2022 ), and these may be new targets for the treatment of HPV-associated CC.…”

“…Among all HPV types in China, the infection rate of HPV52 was relatively low in the late 1990s but has gradually increased since then [ 6 , 7 , 22 ]. Recently, HPV52 has become the most prevalent type among the Chinese population [ 6 , 8 , 9 , 17 19 , 30 ]. Furthermore, the oncogenicity of HPV is mainly attributable to the combined effects of the E6 and E7 oncoproteins.…”

“…The primer sequences specific for the E6 and E7 genes of HPV52 were 52E6E7_F 5′-ACCCACAACCACTTTTTTTTAT-3’ and 52E6E7_R 5′-TTGCCTCTACTTCAAACCAGCC-3’, which were cited from a recently published article [ 8 ]. Primers were synthesized by BGI company (Hangzhou, China).…”
