The genetic response to short-term interventions affecting cardiovascular function: Rationale and design of the Heredity and Phenotype Intervention (HAPI) Heart Study

Abstract: Background-The etiology of cardiovascular disease (CVD) is multifactorial. Efforts to identify genes influencing CVD risk have met with limited success to date, likely due to the small effect sizes of common CVD risk alleles and the presence of gene by gene and gene by environment interactions.

“…HAPI Heart Study. This study was initiated in 2002 to measure the cardiovascular response to 4 short-term interventions affecting cardiovascular health and to identify the genetic and environmental determinants of these responses (10). Before any intervention, baseline BP was obtained manually with the subject in the sitting position after he or she had been sitting quietly for 5 min, and the average of 3 of these measurements was used for analysis.…”

“…21 Confirmation of results was sought in the Framingham Heart Study Offspring Cohort (FHS) 22 and in the cross-sectional Heredity and Phenotype Intervention (HAPI) Heart Study. 23 Table 1 shows the baseline characteristics of the various study samples. Whereas mean estimated GFR (eGFR) was similar across studies, the proportion of people with at least stage 3 CKD (eGFR Ͻ60 ml/min per 1.73 m 2 ) 24 was 3.0% among ARIC white, 2.8% among ARIC black, 1.5% among HAPI, and 8.2% among FHS participants, the oldest study sample.…”

“…They hypothesized that the variation is due to genetic variation in pathways other than thromboxane synthesis. 165 volunteers from the hereditary and phenotype intervention (HAPI) heart study [8] were given 81 mg of aspirin once a day for 2 weeks. GC-MS and LC-MS were performed on serum samples before and after 2 weeks of aspirin ingestion.…”
